小桐子JcNF-YB3和JcNF-YB10基因的克隆与表达分析

【摘要】:以小桐子(Jatropha curcas)c DNA为模版,克隆了Jc NF-YB3和Jc NF-YB10基因的CDS序列。序列分析表明该两个基因分别包含549和528 bp完全阅读框(ORF),编码182和175个氨基酸。Blast搜索结果及进化分析结果表明,Jc NF-YB3和Jc NFYB10都与油桐(Vernicia fordii)NF-YB家族基因序列一致性最高且亲缘关系最近。定量分析表明,Jc NF-YB3和Jc NF-YB10在小桐子根、茎、叶、花、果皮和种子中都有表达,Jc NF-YB3在根中表达量最高,Jc NF-YB10在种子中表达量最高。两个基因在种子发育期的表达量变化趋势一致,但是Jc NF-YB10在种子整个生长发育期的表达量都要高于Jc NF-YB3。Jc NFYB3和Jc NF-YB10都在Na Cl、PEG、4°C低温和ABA处理后表达量不同程度上调,推测这两个基因在小桐子中参与抗逆反应机制和植物激素信号传导。
[Abstract]:The CDS sequences of Jc NF-YB3 and Jc NF-YB10 genes were cloned using (Jatropha curcas) c DNA as template. Sequence analysis showed that the two genes contained 549 and 528 bp complete reading frame (ORF), encoding 182 and 175 amino acids, respectively. Blast search results and evolutionary analysis showed that, Both Jc NF-YB3 and Jc NFYB10 have the highest consistency with the (Vernicia fordii) NF-YB family of Taulownia and have the closest genetic relationship. Quantitative analysis showed that Jc NF-YB3 and Jc NF-YB10 were expressed in roots, stems, leaves, flowers, pericarp and seeds, Jc NF-YB3 was the highest in roots, and Jc NF-YB10 was the highest in seeds. The change trend of the two genes in the seed development stage was the same, but the expression of Jc NF-YB10 was higher than that of Jc NF-YB3.Jc NFYB3 and Jc NF-YB10 in the whole growth and development stage of seeds, but the expression level of Jc NF-YB10 was higher than that of Jc NF-YB3.Jc NFYB3 and Jc NF-YB10 in Na Cl,PEG,. After 4 掳C hypothermia and ABA treatment, the expression of these two genes was upregulated in varying degrees, which suggested that these two genes were involved in stress resistance response and plant hormone signal transduction in Paulownia chinensis.
