高山离子芥耐冷基因的挖掘

【摘要】:冰缘植物高山离子芥具有极强的抗寒能力,具葶离子芥为其同属近缘种,但抗寒性远不及高山离子芥。本试验以高山离子芥和具葶离子芥植株为材料,对这2种离子芥进行低温处理,并采用c DNA扩增片段长度多态性(c DNA-AFLP)技术对其进行耐冷相关基因筛选,共获得与耐冷相关的28条差异表达片段,其中7条来自具葶离子芥,21条来自高山离子芥,并对这28条差异表达的片段进行生物信息学分析。选取高山离子芥特有的5条差异表达片段(TDFs),采用Realtime RT-PCR进行低温诱导表达验证,结果表明确实是低温诱导的特异表达片段。本试验结果为高山离子芥特有耐冷基因的功能研究及其应用提供了依据。
[Abstract]:Alpine ion mustard has strong cold resistance, and scape ion mustard is a relative species, but its cold resistance is not as good as that of alpine ion mustard. In this experiment, the plants of Alpine ion mustard and scape ion mustard were treated at low temperature, and their cold tolerance genes were screened by c DNA amplified fragment length polymorphism (c DNA-AFLP) technique. A total of 28 differentially expressed fragments related to cold tolerance were obtained, of which 7 were from scape ion mustard and 21 from alpine ion mustard. The 28 differentially expressed fragments were analyzed by bioinformatics. Five differentially expressed fragments (TDFs), were selected for low-temperature induced expression verification by Realtime RT-PCR. The results showed that they were indeed specific expression fragments induced by hypothermia. The results of this study provide a basis for the functional study and application of specific cold tolerance genes in Alpine ion mustard.
