胡杨F-box基因克隆和功能分析
[Abstract]:F-box protein can specifically recognize substrate proteins, regulate the signal transduction pathways of many hormones in the ubiquitin proteasome pathway, and respond to various abiotic stresses and participate in plant stress resistance pathway. Populus euphratica (Populus euphratica) is the only tall tree species in northwest desert of China, which has strong resistance to salt and alkali, drought and extreme temperature, and is a candidate tree species for studying the mechanism of forest stress resistance. In order to study the functional characteristics of F-box gene of Populus euphratica, the F-box gene obtained from salt-tolerance correlation analysis was cloned, expressed, and transformed into protoplasts of Populus euphratica. The total length of F-box gene of Populus euphratica is 1 086 bp, encoding 361 amino acids, which contains a F-box domain and a F-box associated domain.q RT-PCR indicating that the F-box gene of Populus euphratica is expressed in all tissues without tissue specificity, and its expression is salt and high temperature. The transient expression of PEG mediated protoplast of Populus euphratica showed that the F-box protein was located on the nucleus, and the overexpression of F-box gene could improve the salt-tolerance of Populus euphratica protoplast by flow cytometry. The study revealed the characteristics of F-box gene expression and salt tolerance of Populus euphratica, which could help to further analyze the function of F-box gene and the molecular mechanism of stress resistance of Populus euphratica.
