含钒蛋黄粉对大鼠生长、氧化应激状态及其相关基因表达的影响

【摘要】:钒是一种重金属,过量的摄入会造成蛋鸡氧化应激,降低鸡蛋品质并残留到鸡蛋中,影响鸡蛋的安全。本研究主要通过给大鼠饲喂含钒的蛋黄粉,考察其对大鼠生长性能、氧化应激状态及其相关基因表达的影响,为评价钒(有机钒)的生物安全性提供依据。选用27只4周龄雌性Wistar大鼠单笼饲养,设3个处理,每个处理9只,分别在饲粮中添加600 g/kg采食3种钒水平(0、5和10 mg/kg)饲粮的蛋鸡所产鸡蛋制备的蛋黄粉。经实测,3种饲粮分别含钒0.107、0.137和0.164 mg/kg。试验期35 d。结果表明:3个处理大鼠生长性能、器官指数,血浆甘油三酯、丙二醛(MDA)、尿素氮含量及谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,肝脏、肾脏超氧化物歧化酶活性、MDA含量、总抗氧化能力,肝脏ALT、AST活性、谷胱甘肽含量和谷胱甘肽巯基转移酶活性,以及肝脏、肾脏组织结构、肾脏钒残留量均无显著差异(P0.05)。与处理1相比,处理2、3大鼠肝脏醌氧化还原酶1(NQO1)活性显著下降(P0.05),NQO1和核因子E2相关因子2 mRNA相对表达量也显著下降(P0.05)。结果提示:大鼠饲粮中添加600 mg/kg不同钒含量的蛋黄粉对大鼠生长性能和机体氧化还原状态的影响无显著差异,但含钒0.137、0.164 mg/kg可以降低NQO1酶活性并下调抗氧化应答相关基因的表达。
[Abstract]:Vanadium is a heavy metal. Excessive intake of vanadium can lead to oxidative stress, reduce the quality of eggs and leave them in eggs, which will affect the safety of eggs. In order to evaluate the biological safety of vanadium (organic vanadium), the effects of vanadium-containing egg yolk powder on growth performance, oxidative stress state and related gene expression in rats were investigated in this study. Twenty-seven 4-week-old female Wistar rats were raised in a single cage and treated with 3 treatments (9 rats each). The egg yolk powder was fed on three kinds of vanadium (0 5 and 10 mg/kg) diets for 600 g/kg, respectively. The measured results show that the three diets contain 0.107 mg/kg. of vanadium and 0.164 mg/kg. of vanadium, respectively. The trial period was 35 days. The results showed that the growth performance, organ index, plasma triglyceride, malondialdehyde (MDA) (MDA), urea nitrogen content, glutamate-alanine aminotransferase (ALT),) -aminotransferase (AST) activity, liver and kidney superoxide dismutase (SOD) activity were measured in three groups. There was no significant difference in MDA content, total antioxidant capacity, liver ALT,AST activity, glutathione content and glutathione mercaptotransferase activity, as well as liver, kidney tissue structure and kidney vanadium residue (P0.05). Compared with treatment 1, the activity of quinone redox enzyme 1 (NQO1) and the relative expression of NQO1 and E2 related factor 2 mRNA were significantly decreased (P0.05). The results showed that there was no significant difference in growth performance and redox state of rats after adding 600 mg/kg different vanadium content of egg yolk powder in diet. However, vanadium 0.137 and 0.164 mg/kg could decrease the activity of NQO1 and down-regulate the expression of genes related to antioxidant response.
