擎天凤梨SAMs基因的分离及开花期的表达分析
[Abstract]:In order to obtain SAMs gene and clarify its role in ethylene biosynthesis, two SAMs genes were obtained by Primer Walking sequencing on the basis of the construction of full-length c DNA library and batch EST sequencing. They were named SAMs1 and SAMs2.SAMs1 gene (Gen Bank:KF787113), respectively. The open reading frame 1086 bp, encodes a 361 amino acid sequence. The sequence of SAMs2 gene (Gen Bank:KF787114) is 1 235 BP, and open reading frame 945 bp, encodes a 314 amino acid sequence. The amino acid composition, physicochemical properties, conserved domain, secondary structure and tertiary crystal structure of two gene encoded proteins were predicted and analyzed by bioinformatics. Molecular phylogenetic analysis showed that SAMs1 was closely related to SAMs gene of Gramineae plants and had a far relationship with SAMs2. The analysis of the expression changes of two genes at anthesis and ethylene release showed that the SAMs1 gene expression was consistent with the change trend of endogenous ethylene release level, that is, the content of SAMs1 gene was the highest in the completely colored red bracts, followed by the green bracts. Finally, green leaves. It is suggested that SAMs1 plays an important role in ethylene biosynthesis, and SAMs2 is involved in the biosynthesis of SAM except ethylene biosynthesis. The results of this experiment are of great significance for the study of ethylene biosynthesis, the color mechanism of bracts and the application of flower promotion.
