花生MAPKKK基因的鉴定和盐胁迫响应分析

【摘要】:MAPKKK(mitogen-activated protein kinase kinase kinase)在植物抗逆胁迫中起重要作用。为分析花生中MAPKKK基因的情况,本试验对耐盐花生(Arachis hypogaea)突变体(S2)和对照(S4)的叶片进行RNA测序分析,筛选出了19个具有完整开放阅读框的MAPKKK基因。它们位于花生A组野生种的8条染色体上,不同成员的氨基酸残基个数、外显子数目差异很大。聚类分析表明19个花生MAPKKK基因能聚到MEKK和Raf两个亚家族。我们利用S2和S4构建了盐胁迫处理前后的表达谱,筛选出4个受盐胁迫诱导表达的MAPKKK基因,分别属于MEKK亚家族(3个)和Raf亚家族(1个)。该研究为花生MAPKKK基因的功能研究和利用提供参考。
[Abstract]:MAPKKK (mitogen-activated protein kinase) plays an important role in plant stress resistance. In order to analyze the situation of MAPKKK gene in peanut, 19 MAPKKK genes with complete open reading frame were screened by RNA sequencing in leaves of salt-tolerant peanut (Arachis hypogaea) mutant (S2) and control (S4). They are located on 8 chromosomes of group A wild species of peanut. The number of amino acid residues and exons of different members vary greatly. Cluster analysis showed that 19 peanut MAPKKK genes could be clustered into two subfamilies of MEKK and Raf. We constructed the expression profiles before and after salt stress with S2 and S4, and screened out four MAPKKK genes induced by salt stress, belonging to MEKK subfamily (3) and Raf subfamily (1) respectively. This study provides a reference for the functional study and utilization of peanut MAPKKK gene.
