扁桃AcCBF2基因的克隆及其在逆境胁迫下的表达分析

【摘要】:本研究根据扁桃生物信息学数据库,采用PCR的方法从扁桃(Amygdalus communis L.)中克隆了一个CBF转录因子基因,命名AcCBF2,基因开放阅读框(open reading frame,ORF)为717 bp,编码238个氨基酸,预测其分子量26.59 kD,等电点为5.29。氨基酸多重序列比对显示,AcCBF2基因编码的氨基酸与其它植物冷胁迫相关的CBF氨基酸序列具有高度同源性,含有一个AP2功能结构域和2个特征基序;系统发育树分析显示,AcCBF2基因属于DREB家族中的A-1亚族,荧光定量显示,AcCBF2基因只对低温和干旱胁迫有响应,对盐胁迫和ABA(脱落酸)没有明显响应。在低温胁迫下,表达量上调;而在干旱胁迫下,表达量先上调后下调。初步预测AcCBF2基因可能对扁桃非生物胁迫有重要的调控作用。
[Abstract]:In this study, according to the bioinformatics database of almond, (Amygdalus communis L.) of almond was obtained by using PCR method. An CBF transcription factor gene was cloned and named AcCBF2, open reading frame (open reading frame,ORF) encoding 238 amino acids for 717 bp,. The predicted molecular weight was 26.59 kD, isoelectric point (5.29). Amino acid multiple sequence alignment showed that the amino acids encoded by AcCBF2 gene had high homology with other plant CBF amino acids related to cold stress, and contained one AP2 functional domain and two characteristic motifs. Phylogenetic tree analysis showed that AcCBF2 gene belonged to A-1 subfamily of DREB family. Fluorescence quantitative analysis showed that AcCBF2 gene was only responsive to low temperature and drought stress, but not to salt stress and ABA (abscisic acid). Under low temperature stress, the expression level was up-regulated, while under drought stress, the expression was up-regulated and then down-regulated. It is preliminarily predicted that AcCBF2 gene may play an important role in the regulation of abiotic stress in almond.
