连翘苷对ApoE基因敲除小鼠主动脉斑块形成的干预作用研究

【摘要】:目的评价连翘苷对主动脉斑块形成的干预作用。方法采用Apo E-/-小鼠喂养西方膳食饲料的方法建立动脉粥样硬化模型,建模同时用连翘苷低、中、高三个剂量(5 mg·kg-1、15 mg·kg-1、45 mg·kg-1BW)进行灌胃干预,连续12周。12周后,比较各组主动脉斑块面积以及Ac LDL刺激下的骨髓来源巨噬细胞PPAR-r、ABCA1、ABCG1、SR-B1基因的表达情况。结果与模型对照组相比,连翘苷中、高剂量组的主动脉斑块面积减少(P0.05),骨髓来源巨噬细胞PPAR-r、ABCA1、ABCG1、SR-B1基因表达均上调(P0.01,P0.05)。结论连翘苷具有一定的抗动脉粥样硬化作用,其机制可能与促进巨噬细胞胆固醇流出的途径有关。
[Abstract]:Objective to evaluate the effect of suspensin on aortic plaque formation. Methods the model of atherosclerosis was established by feeding Apo E-r-mice with western dietary diet. The model was treated with three doses (5 mg kg-1,15 mg kg-1,45 mg kg-1BW) of low, middle and high levels of forsythia glucoside (5 mg kg-1,15 mg kg-1,45 mg kg-1BW). After 12 weeks, the area of aortic plaques and the expression of PPAR-r,ABCA1,ABCG1,SR-B1 gene in bone marrow-derived macrophages stimulated by Ac LDL were compared. Results compared with the model control group, the plaque area of aorta was decreased in the high dose group (P0.05), and the expression of PPAR-r,ABCA1,ABCG1,SR-B1 gene was up-regulated in the macrophages derived from bone marrow (P0.01P05). Conclusion Forsypiin has anti-atherosclerotic effect, and its mechanism may be related to the pathway of promoting cholesterol efflux of macrophages.
