RNA干扰A20基因的表达对人乳腺癌细胞MCF-7增殖、凋亡和迁移的影响

【摘要】:目的利用RNA干扰技术靶向沉默A20基因,对MCF-7细胞株增殖、凋亡和迁移的影响进行研究,探讨A20基因作为乳腺癌治疗靶点的潜在价值。方法人工合成靶向A20基因的siRNA序列和阴性对照(NC)siRNA序列,利用脂质体转染技术将siRNA导入MCF-7细胞株,采用CCK-8细胞增殖实验、Annexin V、7-AAD双染流式细胞术检测凋亡实验和Transwell细胞迁移实验研究沉默A20基因对MCF-7细胞增殖、凋亡和迁移的影响。结果靶向沉默A20基因表达能够有效抑制MCF-7细胞的增殖和迁移,促进细胞凋亡的发生。结论 A20基因在MCF-7细胞的增殖、凋亡和迁移过程中起重要作用,A20基因可能是针对乳腺癌抗肿瘤靶向治疗的潜在靶点。
[Abstract]:Aim to study the effects of RNA interference targeted silencing A20 gene on proliferation, apoptosis and migration of MCF-7 cell line, and to explore the potential value of A20 gene as a therapeutic target for breast cancer. Methods siRNA sequence of targeted A20 gene and (NC) siRNA sequence of negative control were synthesized. SiRNA was transfected into MCF-7 cell line by lipofectamine transfection technique. CCK-8 cell proliferation test, Annexin V, was used. The effects of silencing A20 gene on the proliferation, apoptosis and migration of MCF-7 cells were studied by 7-AAD double staining flow cytometry and Transwell cell migration assay. Results targeted silencing of A20 gene expression could effectively inhibit the proliferation and migration of MCF-7 cells and promote the occurrence of apoptosis. Conclusion A20 gene plays an important role in the proliferation, apoptosis and migration of MCF-7 cells. A20 gene may be a potential target for anti-tumor therapy of breast cancer.
