烟草钾离子通道基因NtTPK的克隆及表达分析

【摘要】:采用同源克隆的方法对烟草品种K326中的一个钾离子通道基因NtTPK进行了研究,该基因序列全长1 317 bp,编码438个氨基酸。实时荧光定量PCR结果表明,该基因在茎中的表达量最高;在低钾处理24 h后该基因表达量最低;200 mmol·L~(-1)NaCl处理24 h后该基因表达量最高;在烟草打顶7 d后,该基因在叶和根中的表达量达到最高。这些结果为进一步研究该基因的功能提供了一定理论基础。
[Abstract]:A potassium channel gene NtTPK in tobacco variety K326 was studied by homologous cloning method. The sequence of the gene was 1 317 bp, encoding 438 amino acids. The results of real-time fluorescence quantitative PCR showed that the expression of the gene was the highest in stem, the lowest after 24 hours of low potassium treatment, and the highest after treatment of 200 mmol 路L ~ (- 1) NaCl for 24 hours, and the expression of the gene was the highest after treatment with 200 mmol 路L ~ (- 1) NaCl for 24 h. After 7 days of tobacco topping, the expression of the gene reached the highest level in leaves and roots. These results provide a theoretical basis for further study of the function of the gene.
