CRISPR-dCas9系统在基因表达调控中的最新研究进展

【摘要】:CRISPR-Cas9系统自问世以来一直被广泛地应用于基因组编辑技术中,直到人们通过点突变的方式将Cas9的Ruv C和HNH剪切结构域失活得到d Cas9后,这个系统又有了新的功能——调节基因的表达水平。研究者通过改变g RNA的结构,并与RNA结合蛋白配对,从而招募转录因子以调节基因的转录水平,达到影响基因表达的目的。本文主要讨论了CRISPR-d Cas9系统的结构特点、作用原理和目前对调节基因表达的最新研究进展,以期为此领域的研究者提供参考。
[Abstract]:Since its inception, CRISPR-Cas9 system has been widely used in genome editing technology, until people inactivated the Ruv C and HNH splicing domains of Cas9 by point mutation to obtain d Cas9. The system has a new function-regulating the level of gene expression. By altering the structure of g-RNA and pairing with RNA-binding proteins, the researchers recruited transcription factors to regulate the transcriptional level of the gene to influence gene expression. In this paper, we mainly discuss the structure and mechanism of CRISPR-d Cas9 system, as well as the latest research progress in regulating gene expression, so as to provide references for researchers in this field.
