马尾松PmCY P77A 1基因的克隆及表达分析

【摘要】:在进行马尾松抗虫材料转录组测序分析时发现一个编码CYP77A1的cDNA,命名为PmCYP77A1。序列分析结果显示,该基因全长1 620 bp,编码539个氨基酸。氨基酸序列分析表明其具有CYP450家族基因的保守结构域,但CYP77A1基因中的FXXGXXXCXG保守特征序列最后一个碱基被A碱基所代替。与其他植物核酸序列进行比对覆盖率和同源性均较差,而氨基酸序列比对覆盖率达到88%~96%,但同源性不高,基本都在50%~55%之间,PmCYP77A1与白云杉遗传聚类最近,与其他被子植物遗传聚类较远。利用荧光定量PCR技术分析得知,PmCYP77A1在马尾松的不同组织中均有表达,其中在嫩茎、雌雄花和嫩叶中的表达量较高,而在球果、成熟组织和根系中的表达量最低。在不同抗虫材料日变化中PmCYP77A1表达量出现先升高后降低的表达趋势,该基因在抗虫材料中表达启动早。研究结果将为进一步研究PmCYP77A1的生物学功能及在马尾松抗虫防御中的作用机理奠定基础。
[Abstract]:In the sequencing analysis of Pinus massoniana insect-resistant material transcriptional group, it was found that a cDNA, encoding CYP77A1 was named PmCYP77A1.. The results of sequence analysis showed that the full length of the gene was 1620 bp, encoding 539 amino acids. Amino acid sequence analysis showed that it had the conserved domain of CYP450 family gene, but the last base of FXXGXXXCXG conserved characteristic sequence in CYP77A1 gene was replaced by A base.
