利用玉米乙酰乳酸合成酶基因ALS点突变处理培育抗除草剂高粱

【摘要】:高粱是重要五大粮食作物之一,因其对很多化学除草剂相当敏感,喷施时需掌握一定的技巧,一般农户不易把握,而培育抗除草剂高粱是重要的途径之一。本研究克隆了玉米品种B73的2个乙酰乳酸合成酶(ALS)基因,进行生物信息学分析高粱、玉米、水稻和拟南芥的ALS基因进化关系、保守结构域以及保守序列,然后进行单、双人工点突变处理ALS基因,最后构建单双点突变超表达载体转化高粱。尽管转化高粱失败,本研究也为进一步培育抗除草剂高粱育种新材料奠定了良好的基础。
[Abstract]:Sorghum is one of the five important food crops, because it is very sensitive to many chemical herbicides, it is necessary to master certain skills when spraying, and it is difficult for farmers to grasp, and cultivating herbicide resistant sorghum is one of the important ways. In this study, two acetyllactic acid synthase (ALS) genes of corn variety B73 were cloned, and the evolutionary relationship, conserved domains and conserved sequences of ALS gene in sorghum, corn, rice and Arabidopsis thaliana were analyzed by bioinformatics. ALS gene was treated with double point mutation, and finally a single and double point mutation overexpression vector was constructed to transform sorghum. Although the transformation of sorghum failed, this study also laid a good foundation for the further cultivation of new herbicide resistant sorghum breeding materials.
