FoxO1及自噬相关基因Beclin1和MAPLC3在胃癌组织的表达及意义

【摘要】:目的探讨叉头转录因子O亚家族1(FoxO1)及自噬相关基因Beclin1和微管相关蛋白轻链3(MAPLC3)在胃癌组织的表达及意义。方法收集本院2015年1月至2016年6月收治的82例手术切除的胃癌组织(胃癌组)及67例癌旁正常组织(对照组),采用荧光实时定量PCR(qPCR)法检测FoxO1、Beclin1及MAPLC3水平,比较组间的水平变化并分析胃癌组中FoxO1、Beclin1及MAPLC3表达变化的相关性,分析组织FoxO1、Beclin1及MAPLC3水平变化与胃癌临床病理参数[性别、年龄、TNM分期、分化程度、部位、淋巴结转移及癌胚抗原(CEA)水平]的关系。结果qPCR检测结果显示,胃癌组的FoxO1、Beclin1及MAPLC3水平依次为(0.489±0.032)、(0.148±0.012)和(0.581±0.038),均低于对照组,差异有统计学意义(P0.05);胃癌组的FoxO1水平与Beclin1水平呈正相关(r=0.270,P=0.014),Beclin1水平与MAPLC3水平呈正相关(r=0.351,P=0.001),但FoxO1水平与MAPLC3水平无关(r=0.208,P=0.061);FoxO1表达与TNM分期、分化程度及淋巴结转移均有关,Beclin1表达与TNM分期、淋巴结转移及CEA水平均有关,且MAPLC3表达与TNM分期、分化程度及CEA水平均有关。结论FoxO1、Beclin1及MAPLC3在胃癌组织中表达降低,且均与TNM分期有关,对胃癌诊断及病情评估有一定价值。
[Abstract]:Objective to investigate the expression and significance of fork transcription factor O subfamily 1 (FoxO1), autophagy related gene Beclin1 and microtubule associated protein light chain 3 (MAPLC3) in gastric cancer. Methods from January 2015 to June 2016, 82 cases of gastric cancer (gastric cancer group) and 67 cases of paracancerous normal tissues (control group) were collected. The levels of FoxO1,Beclin1 and MAPLC3 were detected by fluorescence real-time quantitative PCR (qPCR). The changes of FoxO1,Beclin1 and MAPLC3 expression in gastric cancer group were compared, and the changes of FoxO1,Beclin1 and MAPLC3 levels in gastric cancer group were analyzed. The clinicopathological parameters of gastric cancer [sex, age, TNM stage, degree of differentiation, location) were analyzed.
