水稻细胞质雄性不育恢复基因Rf6功能标记开发及微核心种质中Rf6基因的筛选

【摘要】:核恢复基因Rf6是红莲型杂交水稻特有的新恢复基因,为了寻找更多具有Rf6恢复基因的种质,以扩展红莲型杂交水稻的恢复系来源,组配更多优良杂交品系,利用Rf6及其等位基因的序列差异,开发Rf6基因的功能标记,并利用该标记对中国水稻微核心种质进行PCR扩增,筛选具有Rf6恢复基因的种质。结果表明,在324 bp插入片段(Rf6较其等位基因插入324 bp)两侧设计引物(正向引物:5'-ATGACAAGAGGACCAGCGATGCAATGG-3',反向引物:5'-ATTCTTGCAGAGATAGACCATGAGCGTG-3'),利用该引物可扩增出条带清晰且无杂带的差异片段,可用于Rf6基因的鉴定。利用该标记在197份中国水稻微核心种质中筛选出22份具有Rf6恢复基因的种质,可为拓宽红莲型杂交水稻的恢复系来源提供材料。
[Abstract]:The nuclear restorer gene Rf6 is a unique new restorer gene in red lotus hybrid rice. In order to find more collections with Rf6 restoring gene, to expand the source of restorer lines of red lotus hybrid rice, and to combine more excellent hybrid lines, the nuclear restorer gene Rf6 is a unique new restorer gene in red lotus hybrid rice (Oryza sativa L.). Based on the sequence difference of Rf6 and its alleles, the functional marker of Rf6 gene was developed, and the microcore germplasm of Chinese rice was amplified by PCR, and the germline with Rf6 restorer gene was screened.
