中国水仙凝集素基因在大花烟草中的定量表达及抗蚜性分析

【摘要】:为了确定中国水仙凝集素基因NTL1的抗蚜功能及培育具有抗蚜功能的大花烟草,利用qPCR方法,研究了中国水仙凝集素基因(NTL1)在转基因大花烟草4个株系中的相对表达情况;利用自然感蚜、活体接种和离体接种方法研究了不同转基因株系的抗蚜能力。结果表明,NTL1在不同株系中的相对表达量差异显著,从高到低依次为3,4,1,2;抗蚜虫试验结果显示:转基因烟草具有一定抗蚜性,但不同烟草株系的抗蚜能力有所不同;活体转基因烟草单株与离体烟草叶片的平均蚜虫密度抑制率分别为8.22%~76.61%,4.62%~65.65%;转基因大花烟草可加速蚜虫的死亡。对比NTL1在不同转基因大花烟草株系中的相对表达量及抗蚜效果,发现NTL1在不同株系中的相对表达量与植株抗蚜性具有高度一致性。
[Abstract]:In order to determine the aphid resistance function of Chinese narcissus lectin gene NTL1 and to cultivate large flower tobacco with aphid resistance, the relative expression of Chinese narcissus lectin gene (NTL1) in four transgenic tobacco lines was studied by qPCR, and the aphid resistance of different transgenic lines was studied by natural aphids, in vivo and in vitro. The results showed that the relative expression of NTL1 in different lines was significantly different, from high to low was 3, 4, 1, 2. The results of aphid resistance test showed that transgenic tobacco had certain aphid resistance, but the aphid resistance ability of different tobacco lines was different, and the average aphid density inhibition rates of single transgenic tobacco and in vitro tobacco leaves were 8.22% 76.61% and 4.62% 65.65%, respectively. the results showed that transgenic tobacco had certain aphid resistance, but the average aphid density inhibition rate of transgenic tobacco was 8.22% 76.61% and 4.62% 65.65%, respectively. Transgenic tobacco can accelerate the death of aphids.
