肝细胞癌差异基因分析及长链非编码RNA的初步验证 ——基于转录组芯片的研究

研究目的和意义肝细胞癌(Hepatocellular carcinoma,HCC)是世界上高发的恶性肿瘤之一,其死亡率居恶性肿瘤的第三位。在中国,肝细胞癌的新发病例数和死亡率高居世界前列,其中,乙型肝炎病毒感染占大多数。在广西地区,肝细胞癌是死亡率最高的恶性肿瘤。由于肝细胞癌起病隐匿、肿瘤生长迅速、转移性高,多数患者就诊时已经属于中晚期。虽然目前肝脏手术切除或肝移植等治疗手段可缓解病人情况,但是术后肿瘤复发和转移严重影响患者的生活质量。因此,深入研究肝细胞癌的发生、发展机制,探索针对肝细胞癌早期、特异性诊断标志物和治疗靶点十分必要,这将为肝细胞癌的诊断和治疗提供有力依据。肝细胞癌是一种高度异质性的基因源性肿瘤。肿瘤基因本身不仅可导致细胞癌变,而且可通过表观遗传学水平调控引起肝细胞的癌变,主要包括甲基化、乙酰化、基因印记、染色体修饰和非编码RNA调控等。其中,最近的研究证实,长链非编码RNA(long noncoding rnas,lnc RNAs)在恶性肿瘤发生、发展中起到重要作用,如lnc RNA PCA,HOTAIR,H19等。长链非编码RNA是一类长度大于200nt核苷酸RNA,...
