苹果树根际高效解钾菌的筛选及鉴定
[Abstract]:In order to find out the population characteristics and properties of kalium lysozyme isolated from apple tree rhizosphere soil, a high potassium hydrolytic bacteria was isolated from the rhizosphere soil of apple tree. In this study, 118 strains with potassium hydrolysis activity were isolated from potassium feldspar as the sole potassium source. The repeat fragment PCR gene fingerprinting (Repetitive-element PCR,rep-PCR) was clustered into 29 groups. Using flame spectrophotometry to determine the ability of potassium hydrolysis of the strain, 5 strains of high efficiency potassium hydrolyzing bacteria were obtained. The average activity of potassium hydrolysis reached 41.47mg L -1, in which K105 had the strongest ability to dissolve potassium, and the available potassium increased by 23.09%. The K concentration measured by H2O2 was increased and the available potassium increased to 31.22. Morphological observation, physiological and biochemical characteristics detection and phylogenetic analysis based on 16s r RNA gene sequence were used to identify the highly efficient Kaliolytic bacteria strains. The results show that K1 is Acinetobacter (Acinetobacter sp.), K98 K105 and K115 is Pseudomonas (Pseudomonas sp.), K168 and Bacillus (Bacillus sp.). The results can provide a basis for the development of a new type of high-efficient potassium hydrolytic bacteria and for the improvement of continuous cropping soil and the development of special microbial fertilizer for apple.
