舞毒蛾高毒力Bt杀虫蛋白及协同增效组合的筛选

【摘要】:舞毒蛾Lymantria dispar(Linnaeus)是世界性的重要农林业害虫。研究Bt蛋白对舞毒蛾幼虫的毒杀作用,筛选出高毒力Bt蛋白及具有协同增效作用的蛋白组合,将为舞毒蛾的生物防治提供依据。本研究提取了十一种Bt蛋白对三日龄舞毒蛾幼虫进行初步筛选,初筛结果显示Cry 1Ac、Cry 2Ab、Cry 1Ca、Vip3Aj四种蛋白具有较高毒力。进而分别测定了这四种蛋白7天的LC50。结果表明,杀虫毒力最高的为Cry1Ac,其LC50达到了2.47 μg/g;Cry3Ab和Cry1Ca的LC50分别为29.19μg/g和42.45μg/g,表现出了较强的杀虫活性;而Vip3Aj的LC50为101.26 μg/g,具有中等毒力,该结果是对Vip3Aj具有杀虫活性的首次报道。进一步将4种蛋白组合(Crr1Ac与Cry2Ab、Cry1Ac与Cry1Ca、Cry1Ac与Vip3Aj、 Cry2Ab与Cry1Ca)按1:1混配,测定蛋白组合对舞毒蛾的杀虫活性和增效作用。结果发现,Cry1Ac+Cry2Ab混配蛋白LC50达到0.64 μg/g,协同毒力指数最高,为7.098,具有很强的协同增效作用;Cry1Ac+Cry1Ca和Cry1Ac+Vip3Aj混配蛋白LC50分别达到0.76μg/g和1.06μg/g,协同毒力指数分别为6.162和4.539,具有显著的增效作用。
[Abstract]:Lymantria dispar (Linnaeus) is an important agricultural and forestry pest in the world. To study the toxic effect of Bt protein on the larvae of Plutella xylostella, and to screen out the high virulence Bt protein and the protein combination with synergistic effect, which will provide the basis for the biological control of Plutella xylostella. In this study, 11 kinds of Bt proteins were extracted to screen the larvae of three-day-old Plutella xylostella. The results showed that Cry 1AC, Cry 2Ab, Cry 1Caand Vip3Aj proteins were highly virulent. Furthermore, the LC50. of these four proteins were determined for 7 days. The results showed that the highest insecticidal toxicity was that the LC50 of Cry1Ac, reached 2.47 渭 g 路g / g and the LC50 of Cry1Ca was 29.19 渭 g / g and 42.45 渭 g / g, respectively, showing strong insecticidal activity, while the LC50 of Vip3Aj was 101.26 渭 g 路g / g, which was the first report of insecticidal activity against Vip3Aj.
