4个Y-STR基因座的多态性及其法医学应用的研究

【摘要】:通过荧光标记引物结合ABI 377型全自动DNA测序仪检测自动分型方法对中国壮族及汉族人群中各 10 0例无关男性个体的A10、C4、A7 1、A7 2等 4个Y染色体特异的基因座的等位基因及单倍型的分布进行了调查。结果发现A10、C4、A7 1、A7 2基因座分别有 7、6、6、6个等位基因 ,基因多样性 (GD)分别为 0 7776 0 6 2 9(壮 汉 )、0 773 0 732、0 5 978 0 72 72、0 6 6 6 4 0 6 4 5 8。在 2 0 0个观察样本中共发现 114种单倍型 (haplotype) ,单倍型多样性 (haplotypediversity,HD)分别为 0 9786 0 9772 (壮 汉 )。通过测序确认基因座核心重复序列及等位基因核心序列重复数。建立了这 4个基因座的复合扩增体系 ,分型准确清晰。还对这 4个基因座的男性特异性、遗传稳定性、灵敏度等法医学有关指标进行了考察并且在实际案例中进行了应用 ,结果证明 ,这 4个Y STRs基因座非常适应于法医检验 ,具有较高的实用价值
[Abstract]:Detection of four Y chromosomal specific loci in 100 unrelated male individuals in Chinese Zhuang nationality and Han nationality by fluorescent marker primer combined with ABI 377 automatic DNA sequencing apparatus for 4 Y chromosomal specific loci, such as A10C _ 4, A _ 4, A _ 7 _ 1, A _ 7 _ 2, etc.The distribution of site gene and haplotype was investigated.The results showed that there were 6 alleles and 6 alleles in A10C4, A7 1 and A7 2 loci, respectively, and the genetic diversity was 0 7776 629 (Chong Hou, 0 773 0 732, 0 5 978 0 72 72 2, 0 66 4 048 8), respectively.A total of 114 haplotype haplotypes were found in 200 observation samples. The haplotype diversity of haplotype was 0 9786 9772.The core repeats of loci and alleles were confirmed by sequencing.The multiplex amplification system of these four loci was established, and the typing was accurate and clear.The male specificity, genetic stability and sensitivity of the four loci were investigated and applied in practical cases.These four Y STRs loci are very suitable for forensic examination and have high practical value.
