新Y-STR基因座DYS714遗传多态性研究

【摘要】: 目的:本文对山西汉族群体Y-STR DYS714基因座的等位基因频率分布进行调查,并对该基因座的法医学应用价值进行探讨。 方法:在GDB数据库中查找该基因座;在重复序列两端设计引物;用PCR方法扩增:电泳分型;对其所有等位基因测序:构建等位基因分型标准物;按照ISFG原则命名等位基因。 结果:本文研究显示:DYS714基因座核心序列为ttttc,被调查的山西地区汉族男性个体核心序列的重复次数为10~14,共5个等位基因:10、11、12、13、14。基因频率分别为0.15、0.1583、0.2167、0.3333、0.1416,基因多样性为0.7808,个人识别能力(DP)和非父排除率(PE)均为0.7808。20例女性样本DNA无扩增产物。同一尸体血液,器官组织检测结果分型一致。15例两代家系观察未见突变。 结论:新筛选到的DYS714具有较高的遗传多态性,在法医学及人类学方面具有应用价值。
[Abstract]:Objective: to investigate the allele frequency distribution of Y-STR DYS714 locus in Shanxi Han population, and to explore the value of forensic application of this locus. Primers were designed at both ends of the repeat sequence, amplified by PCR: electrophoretic typing; all alleles were sequenced; alleles were constructed; Results: according to ISFG principle, the core sequence of WDYS714 gene was ttttc. the number of repeats of core sequence of male Han nationality in Shanxi area was 101.The total number of 5 alleles was 10: 111112C1313140.Results: the alleles were named as alleles according to ISFG principle. The results showed that the core sequence of WDYS714 gene was ttttc. The gene frequencies were 0.1583 (0.2167), 0.3333 (0.1416), and (0.7808), the individual recognition ability (DPDP) and the non-paternal exclusion rate (PEE) were 0.7808.20 female samples, respectively. The results of blood, organ and tissue examination of the same cadaver were consistent. 15 cases of two generations were observed without mutation. Conclusion: the newly selected DYS714 has higher genetic polymorphism and has application value in forensic medicine and anthropology.
