法医鱼类学种属鉴别初步研究

【摘要】:目的 法医鱼类学是一门正在兴起的边缘科学,它是在鱼类学、分子生物学、分析化学、生物信息学等基础上,特别是汲取了法医DNA分析的经验发展起来的,旨在对各类案件涉及到的鱼类样本进行鉴定,为案件的侦查与审判提供科学证据。法医鱼类学在鱼类资源科学管理和保护方面有重要作用。本课题旨在研究用于法医鱼类学种属鉴别的新技术。 方法 选择鲤形目的5种珍稀鱼类作为研究对象,,它们是胭脂鱼,重口裂腹鱼,岩原鲤,华鲮,四川白甲。另外20种鱼作为研究的对照样本。全部样本采自长江、嘉陵江、雅江以及重庆和成都水产品市场。根据GenBank中鲤鱼的DNA序列资料,利用引物设计软件设计鲤形目控制合成5.8SrRNA和28SrRNA基因间的转录间隔子2(Internal Transcribed Spacer 2,ITS2)DNA区域的通用引物,预计扩增片段包含完整的ITS2及部分编码5.8SrRNA和28SrRNA基因的序列。用通用引物对样本进行PCR扩增,PCR产物用聚丙烯酰胺凝胶电泳分离,回收。对每种要鉴别鱼类的回收产物进行DNA双向测序。根据测序结果,用系统进化软件(Mega3.1)进行鲤形目鱼类亲缘关系分析。根据鲤形目5种鱼类ITS2序列的不同,设计分别针对每种鱼的种属特异
[Abstract]:Objective Forensic fish science is an emerging frontier science. It has been developed on the basis of fish science, molecular biology, analytical chemistry, bioinformatics and so on, especially drawing on the experience of forensic DNA analysis. The purpose of this paper is to identify the fish samples involved in various cases and to provide scientific evidence for the investigation and trial of the cases. Forensic fish science plays an important role in the scientific management and protection of fish resources. The purpose of this paper is to study a new technique for the identification of species in forensic fish. Methods five rare fishes of Cyprinidae were selected as research objects. They were cochineal fish, carmine fish, Carpio carpio, mud carp and Sichuan white nail. Another 20 species of fish were used as control samples in the study. All samples were collected from Yangtze River, Jialing River, Yajiang River and aquatic products markets in Chongqing and Chengdu. Based on the DNA sequence of Cyprinus Carpio in GenBank, a general primer was designed to control the synthesis of 5.8s rRNA and 28s rRNA intergenic intertranscriptional spacer 2 (IS2) DNA region by using primer design software. The amplified fragments are expected to contain complete ITS2 and partial sequences encoding 5.8s rRNA and 28s rRNA genes. The PCR products were separated and recovered by polyacrylamide gel electrophoresis. DNA bidirectional sequencing was performed for each recovery product to be identified from fish. Based on the sequencing results, phylogenetic analysis of Cyprinidae was carried out by means of phylogenetic software (Mega 3.1). According to the different ITS2 sequences of 5 species of Cyprinidae, the specific species of each fish were designed.
