豹猫细胞体外培养体系建立及其生物学特性分析

【摘要】:对豹猫6种组织进行原代培养建系,最后选择3种组织来源细胞即剑状软骨源细胞、心脏源细胞和肺源细胞。以该3种细胞为实验材料研究豹猫细胞的生物学特性。在本研究中,主要进行了如下实验:细胞的原代建系及传代培养的形态观察、细胞贴壁率计算、冻存复苏率对比、3种细胞生长曲线的对比、支原体污染检测、核型分析等。在形态学观察方面,3种来源细胞各有显著特征但均属于成纤维样细胞。贴壁率能力分析显示,心脏源细胞最弱;肺源细胞代次P9之前易贴壁,P9之后细胞贴壁增殖及传代比较困难;剑状软骨源细胞最易贴壁。将三种细胞冻存前后存活率进行实验对比发现:解冻后相比于冻存前均有所下降,随着细胞代次的增高,细胞的存活率也有所下降。生长曲线对比发现,曲线均呈"S"型曲线,曲线显示剑状软骨源细胞增殖的最快,肺源细胞次之,心脏源细胞最慢。支原体污染检测细胞污染情况为阴性。在豹猫细胞核型分析研究中发现,染色体数目为2n=38,18对为常染色体,形态类型为7M+1 1SM;1对为性染色体,其形态类型X为ST。本研究为日后利用豹猫细胞、保存遗传信息及深入研究其生物学特性提供了一个好的依据和实验材料。
[Abstract]:Six tissues of leopard cat were cultured and established in primary culture. Finally, three kinds of tissue derived cells, namely, swordlike chondrocytes, heart-derived cells and lung derived cells, were selected. The biological characteristics of leopard cat cells were studied by using these three kinds of cells as experimental materials. In this study, the following experiments were carried out: morphological observation of primary cell line and passage culture, calculation of cell adhesion rate, comparison of three cell growth curves, detection of mycoplasma contamination, karyotype analysis, etc. In the aspect of morphological observation, the three kinds of cells have distinct characteristics but all belong to fibroblast-like cells. The ability of adherent rate analysis showed that the heart derived cells were the weakest, the lung derived cells were easy to adhere to P9 before the passage of P9 and it was difficult to adhere to the proliferation and passage of the cells after P9, and the sword chondrocytes were the most likely to adhere to the wall. The survival rate of three kinds of cells before and after cryopreservation was compared with that before thawing, and the survival rate decreased with the increase of cell generation. The growth curves were found to be "S" curve, which showed the fastest proliferation of chondrocytes, followed by lung cells and heart cells. Mycoplasma contamination was negative for cell contamination. In the analysis of the karyotype of leopard cats, it was found that the number of chromosomes was 2nrn ~ (38), 18 pairs were autosomes, and 7M _ (11) SMN _ (1) pairs were sex chromosomes, and its morphological type X was ST. This study provides a good basis and experimental material for the future use of leopard cat cells, the preservation of genetic information and the further study of its biological characteristics.
