甜瓜MADS-box基因家族成员的鉴定及CmMADS02和CmMADS26基因的功能分析

【摘要】：甜瓜(Cucumis melo L.)是一年生双子叶的草本蔓生植物,也是一种重要的园艺作物和营养丰富的水果,被广泛种植于世界各地并誉为世界十大水果之一。甜瓜是典型的雄花和两性花同体植物,其花器官发育明显符合ABC(D)E发育模式。本实验所用材料是典型的呼吸跃变型果实,是研究鲜果果实发育成熟的理想对象。MADS-box基因家族编码一类转录因子,对植物生长发育,尤其是对生殖器官发育的调控起着重要的作用,是花发育及果实成熟的重要调控因子。本论文以甜瓜品种河套蜜瓜(Cucumis melo L.cv.Hetao)为实验材料,应用生物信息学的方法从其基因组中鉴定筛选出MADS-box基因家族的全部成员,共鉴定得到62个甜瓜MADS-box基因,其中26个属于Ⅰ型基因,分布在Mα、Mγ和Mδ亚家族中;36个属于Ⅱ型基因,分布在MIKCc型各亚家族中。其中,在Mβ、AGL12、BS、TM8和MIKC*亚家族中没有任何基因的分布。同时对该家族所有基因进行了染色体定位、内含子-外显子结构、保守基序分布、系统发生与进化关系分析,结果显示,大部分成员的特性均与其所在亚家族的其他成员相一致,这也说明在不同物种间,MADS-box家族基因在结构和进化上是相对保守的。应用半定量方法对甜瓜MADS-box基因家族各成员进行了表达分析,根据检测结果,从每个亚家族选取1～2个表达量较显著且表达模式有特点的基因,采用实时荧光定量法进一步对其表达特性进行检测,结果显示,不同亚家族基因呈现出不同的表达模式,预示着其广泛的生物学功能。应用RT-PCR法克隆了CmMADS02和CmMADS26基因全长cDNA,并构建其超表达和RNAi载体。应用瞬时表达技术分析其在甜瓜果实成熟中的作用,结果显示CmMADS26基因对甜瓜果实成熟可能具有促进作用。对转CmMADS02基因的T1代甜瓜植株表型分析显示,CmMADS02基因可能参与调控甜瓜两性花雄蕊的发育。
[Abstract]:Cucumis melo L. It is an annual dicotyledonous herbaceous vine. It is also an important horticultural crop and nutritious fruit. It is widely planted all over the world and is regarded as one of the top ten fruits in the world. Muskmelon is a typical male flower and hermaphrodite plant, its floral organ development obviously accords with ABC(D)E development pattern. The material used in this study is a typical respiratory variant fruit. It is an ideal target for studying the development and maturation of fresh fruit. MADS-box gene family encodes a class of transcription factors, which is useful for the growth and development of plants. Especially, it plays an important role in regulating the development of reproductive organs, and is an important regulatory factor for flower development and fruit maturation. In this paper, using Cucumis melo L.cv. Hetao, a melon variety, as the experimental material, all members of the MADS-box gene family were identified by bioinformatics method. A total of 62 MADS-box genes of muskmelon were identified, of which 26 belonged to type 鈪，
