转录因子和染色质特征的分布特点及其关联

【摘要】:基因表达的调控涉及转录因子、组蛋白修饰和其他因素之间复杂的相互作用。转录因子和表观遗传修饰共同调节基因的表达,并在其中起到重要的作用,同时有研究表明,转录因子和表观遗传修饰之间有着密切联系。表观遗传修饰和转录因子之间的研究表明染色质中的某些因素作用于转录因子附近或转录起始位置;染色质特性与共占有转录因子的研究依然还是受到许多因素限制,目前有很多种计算方法来衡量染色质特征与转录因子之间的相关性来鉴定转录因子共占位点,但每种方法都有不同缺陷。本文选择两个人类细胞系,分别是来自ENCODE数据的人类胚胎干细胞和人类宫颈癌细胞,包括单独转录因子和共占有转录因子共80个,研究不同的染色质特征与转录因子的关系。首次提出chromatin state特征,结合染色质特征包括DNase Ⅰ、11种组蛋白修饰、DNA甲基化,利用统计分析的方法预测共占有转录因子,发现H3K27ac、H3K4me2、H3K4me3和H3K9ac是更可靠的预测因子,融合DNA甲基化可以把预测准确率提高2%左右,chromatin state特征在某些转录因子预测中可以达到90%准确率。
[Abstract]:The regulation of gene expression involves the complex interaction between transcription factors, histone modification and other factors. Transcription factors and epigenetic modification jointly regulate the expression of genes and play an important role in them. At the same time, some studies have shown that there is a close relationship between transcription factors and epigenetic modification. The study between apparent genetic modification and transcription factors shows that some factors in chromatin act on the vicinity of transcription factors or transcription initiation position; the study of chromatin characteristics and co-possession transcription factors is still limited by many factors. At present, there are many calculation methods to measure the correlation between chromatin characteristics and transcription factors to identify transcription factor co-occupying sites, but each method has different defects.
