‘蓓蕾’蓝靛果花色苷组成鉴定及其单体微胶囊稳定性的研究
[Abstract]:In this paper, the extraction and purification of anthocyanin from a new breed of "Bud Bud" Lonicera indigo were studied. The samples of anthocyanin freeze-dried powder were prepared by the optimum technology, and the total amount and species of anthocyanin were detected and identified. The antioxidant capacity was compared and analyzed. In addition, the highest content of anthocyanin-3-glucoside of Lonicera edulis was prepared. In order to improve its stability, the microencapsulation of cornflower-3-glucoside was carried out in order to improve its stability, and the stability of high-cornflower-3-glucoside microcapsule was studied. It provides data support for the product promotion of Lonicera edulis and the application of the-3-glucoside monomer of cornflower-3-glucoside. The main results are as follows: (1) the effects of extraction agents, ultrasonic extraction time, ratio of material to liquid and concentration of extractant on the extraction of anthocyanin from 'Bud' Lonicera indigo fruit were studied by single factor test. The optimum technological parameters for extracting anthocyanin from 'Bud Bud' Lonicera edulis fruit were obtained by orthogonal test and variance analysis. The optimum technological parameters were as follows: 60% volume fraction was extracted by 0.1% hydrochloric acid acidified ethanol. The ratio of solid to liquid was 1: 10 mg / mL, and the extraction time was 90 min. (2) in the study of anthocyanin purification, it was found that the adsorption of D101 macroporous resin was the best. Some important technological parameters, such as pH ~ (3), adsorption time of 120 min ~ (60%) and ethanol elution, were determined. Under these conditions, the anthocyanin content in the crude extract of 'Bud' Lonicera edulis was 353.35 卤0.79 mg / g, and the purity was increased from 4.76% to 35.43%. (3) the anthocyanin composition of 'Bud'Lonicera indigo was identified by HPLC-MS, and 11 anthocyanins were found. Among them, the total amount of anthocyanin was 90.679 9%, and the first time was found in Lonicera edulis fruit. In addition, the total antioxidant capacity (T-AOC) and the free radical scavenging ability of ABTS were measured to compare and analyze the antioxidant ability of anthocyanin extract of Lonicera edulis and the VC of Corydanthemum 3-glucoside. The results showed that the order of antioxidant ability of the above three substances was as follows: (4) the highest content of the anthocyanin of 'Bud'Lonicera indigo was prepared by semi-prepared liquid chromatography. The purity of the freeze-dried powder is 97.8%. The optimum conditions for preparation of cornflower -3-glucoside microcapsules were as follows: core material: wall material 1: 97, soy protein isolate: maltodextrin 2: 8, homogenization time 8 min. The results showed that the encapsulation rate was 92.17 卤1.87, the encapsulation rate was 78.42 卤1.23, the solubility was 76.21 卤2.14 and the moisture content was 5.98 卤0.5. (5) the results showed that cornflower -3- glucose was sensitive to light and high temperature, but air had little effect on it. The results showed that the embedding treatment could significantly improve the stability of sunlight, temperature, oxygen and digestion of cornflower-3-glucoside.
