【摘要】:蜂胶是一味中药,是由西方蜜蜂采集植物树脂并混入其上颚腺、蜡腺等分泌物形成的胶黏性物质。蜂胶具有广泛的药理学活性,如抗病原微生物、抗氧化、抗炎、抗肿瘤等,目前被普遍应用于食品和保健品中以用来预防疾病和增进健康。我们前期实验表明,EECP具有很好的抗肿瘤活性,但功效成分及作用机理并不完全清楚,此外,蜂胶不同的提取方法也影响到蜂胶的抗肿瘤活性。基于此,本论文主要研究了(1)EECP和EEBP的抗肿瘤活性比较;(2)在炎症微环境下,EECP及其主要组分CAPE的抗肿瘤功效及作用机理;(3)CPWE的抗肿瘤活性及其功效成分;(4)Ea EP中主要成分的抗肿瘤活性及其功效。研究结果如下:1.相同剂量的EECP比EEBP抑制肿瘤细胞增殖作用更明显。2.在LPS诱导三阴性乳腺癌MDA-MB-231细胞中,EECP及其主要组分CAPE能够显著地抑制乳腺癌细胞的增殖、迁移。进一步研究表明,EECP及其主要组分主要通过下调TLR4的表达,抑制其下游信号分子My D88、IRAK4、TRIF、NF-κB p65和NO的水平,同时上调LC3-Ⅱ和p62的蛋白表达,活化细胞中的凋亡信号分子Caspase 3蛋白,促进LPS诱导的MDA-MB-231细胞的凋亡。3.CPWE共分离得到11种成分(咖啡酸、阿魏酸、异阿魏酸、3,4-二甲氧基肉桂酸、短叶松素、咖啡酸苄酯、CAPE、芹菜素、松属素、白杨素、高良姜素)。其中四种酚酸类化合物对肿瘤细胞没有明显的抑制作用,两种酯类和五种黄酮类均具有很好的抑制肿瘤细胞增殖的作用;进一步研究表明,CPWE中主要抗肿瘤组分通过上调ROS的水平,激活细胞凋亡信号分子Caspase 3蛋白的表达,促进肿瘤细胞的凋亡。4.通过甲醇-水溶液梯度洗脱Ea EP得到I、II、III、IV、V、VI、VII、VIII八组组分。并研究了抗肿瘤活性发现,蜂胶洗脱部分中III、IV、V具有较强的抗肿瘤功效,通过活化Caspase 3,上调Annexin7、LC3-Ⅱ等蛋白的表达,下调p62、4E-BP1、P4E-BP1等蛋白的表达促进肿瘤细胞的凋亡和自噬。
[Abstract]:Propolis is a traditional Chinese medicine, is the western bee collecting plant resin and mixed with their mandibular gland, adhesive substance wax gland secretions to form. Propolis has wide pharmacological activity, such as anti microbial, anti-inflammatory, antioxidant, anti-tumor, is currently widely used in food and health care products in order to prevent disease and improve health. Our previous experiments showed that, EECP has good antitumor activity, but the effectiveness of the composition and mechanism is not entirely clear, in addition, the different extraction methods of propolis can affect the antitumor activity of propolis. Based on this, this paper mainly studies the comparison of EECP (1) and the antitumor activity of EEBP; (2) in the inflammatory microenvironment, antitumor effect and mechanism of EECP and its main components of CAPE; (3) the anti-tumor activity and the effect of the composition of CPWE; (4) the main component of the Ea EP in the anti-tumor activity and efficacy. The results were as follows: 1. the same dose of EECP than EEBP inhibited the proliferation of tumor cells was more obvious in.2. LPS induced three negative breast cancer MDA-MB-231 cells, EECP and its main components of CAPE can significantly inhibit the proliferation of breast cancer cells, migration. Further study showed that EECP and its main components mainly through down regulating the expression of TLR4 and its downstream signal suppression D88 IRAK4, TRIF molecules My, NF-, p65 kappa B and NO levels, and up regulation of LC3- II and p62 protein expression and activation of apoptotic signal molecules in Caspase cells and 3 protein, promote the apoptosis of.3.CPWE induced by LPS in MDA-MB-231 cells were isolated from 11 ingredients (caffeic acid, ferulic acid and isoferulic acid. 3,4- two methoxy cinnamic acid, Ye Songsu short, caffeic acid benzyl ester, CAPE, apigenin, pinocembrin, chrysin, Takara Jiangso). The four kinds of phenolic compounds have no obvious inhibitory effect on tumor cells, two esters and five yellow Ketones have good inhibitory effects on tumor cells; further study showed that CPWE mainly anticancer components through upregulation of ROS expression level, activation of apoptotic signaling molecules Caspase 3 protein, promote the apoptosis of.4. tumor cells by gradient elution of methanol water solution Ea EP I, II, III, IV V, VI, VII, VIII, eight groups of components. And to study the antitumor activity of propolis, was part of III, IV, V has strong anti-tumor effect, through the activation of Caspase 3, up regulation of Annexin7 expression, LC3- II protein down regulated p62,4E-BP1 expression of P4E-BP1 protein in promoting apoptosis and autophagy the tumor cells.
