透明细胞肾细胞癌长链非编码RNA表达谱分析及初步验证

【摘要】:目的探讨及分析透明细胞肾细胞癌(ccRCC)长链非编码RNA(lncRNA)的表达谱,并进行初步验证。方法利用高通量lncRNA芯片技术分别检测3例ccRCC组织及对应癌旁组织中lncRNA表达谱的差异,对芯片结果进行基因本体论(Gene Ontology)分析,Pathway分析,建立lncRNA与靶基因的基因网络图,并利用实时荧光定量RT-PCR技术对高通量lncRNA芯片检测结果进行验证。结果 ccRCC患者癌组织与癌旁对照样本差异表达的lncRNA共3 862条,其中1 619条上调表达,2 243条下调表达(差异表达倍数≥2.0,P0.05)。荧光定量RT-PCR结果显示,筛选出的6个lncRNA,在87例ccRCC样本中的表达差异与芯片检测结果一致。结论ccRCC存在明显的lncRNAs差异性表达,这些lncRNAs及某些信号通路可能在肾癌发生和发展中发挥重要作用。
[Abstract]:Objective to investigate and analyze the long chain noncoding RNA-ln cRNAs expression profile of clear cell renal cell carcinoma (RCC). Methods High throughput lncRNA microarray technique was used to detect the difference of lncRNA expression profiles in 3 cases of ccRCC and corresponding adjacent tissues.
