亚抑菌浓度替加环素对鲍曼不动杆菌生物膜影响的研究

【摘要】:目的:探讨亚抑菌浓度替加环素对多重耐药鲍曼不动杆菌生物膜的影响。方法:用微量肉汤稀释法测得替加环素的最低抑菌浓度(MIC),然后同时做a、b两组实验,a组先体外构建鲍曼不动杆菌生物膜的模型,待其形成成熟的生物膜后,再用替加环素破坏成熟生物膜;b组为替加环素抑制鲍曼不动杆菌生物膜的实验,两组实验均设立实验组和对照组。a组中实验组为Ⅰ、Ⅱ、Ⅲ组,对照组为Ⅳ组。实验组Ⅰ、Ⅱ、Ⅲ组中分别用MIC、1/2MIC、1/4MIC亚抑菌浓度的替加环素作用于已形成的成熟的生物膜,48h、96h后取出;对照组Ⅳ组不加替加环素,等量TSB培养基培养细菌。通过结晶紫半定量染色法对生物膜进行染色,共聚焦倒置显微镜下观察生物膜染色情况,并采用多功能酶标仪在570nm波长处测得生物膜OD值;b组中设立A、B、C三个实验组和D组对照组。A、B、C三个实验组分别用MIC、1/2MIC、1/4MIC亚抑菌浓度的替加环素培养液培养MDR-Ab,对照组D组中则只用TSB培养基培养,不加替加环素。相同方法测定生物膜OD值及镜下观察生物膜形态变化。结果:a组中替加环素在实验组Ⅰ、Ⅱ、Ⅲ组中,即MIC、1/2MIC、1/4MIC浓度下作用于已成熟生物膜48h、96h后,所测得OD值与对照组OD值之间进行比较,结果统计学有显著性差异(P0.05);b组三个实验组A、B、C中所形成生物膜测得的OD值与对照组D组中OD值比较均有统计学差异(P0.05),且A、B组所测OD值小于D组中OD值,C组实验组中OD值大于D组中OD值。结论:替加环素在亚抑菌浓度下具有破坏MDR-Ab成熟生物膜的能力;MIC、1/2MIC浓度下的替加环素培养液可以抑制鲍曼不动杆菌生物膜的形成,而1/4MIC浓度下的替加环素培养液诱导生物膜的形成。
[Abstract]:Objective: to investigate the effect of tegacyclin on the biofilm of Acinetobacter baumannii multidrug resistance. Methods: the minimal inhibitory concentration of tegicyclin was determined by broth dilution method, and then the two groups were treated as group A at the same time.
