川产黄连及其炮制品降糖化学成分与药效研究

【摘要】:黄连为毛茛科植物黄连Coptischinensis Franch、三角叶黄连Coptis deltoidea C.Y.Cheng et Hsiao.或云连Coptis teeta Wall.的干燥根茎。秋季采挖,除去须根及泥沙,干燥,撞去残留须根。黄连因基源不同,药材分为"味连"、"雅连"和"云连"。黄连是一味川产道地药材,四川是其主产区,主要有味连、雅连,另有四川历史习用品种峨眉野连 Coptis omeiensis(Chen)C.Y.Cheng.(习称"凤尾连")。(1)首先建立了同时测定7种黄连生物碱成分的HPLC含量测定方法,经方法学验证该方法准确性、重现性、精密度、稳定性、线性均符合要求。测定6批味连、6批雅连、3批凤尾连的7种生物碱的含量以及总生物碱的含量,对比不同种之间总生物碱以及7种生物碱分别的含量差异,并分析3个品种之间化学成分的差异。结果表明:味连的总生物碱含量最高,7种生物碱在味连、雅连、凤尾连中含量各不相同,差异较大。(2)通过四因素三水平的正交实验,以生物碱总含量为指标,筛选了酒黄连、姜黄连、萸黄连的最佳炮制工艺;以雅连为例,对比了黄连不同炮制品与生品的总生物碱含量以及7种不同生物碱各自的含量差异。结果表明,萸黄连炮制后总生物碱含量最高,且高于生品,姜黄连次之,亦高于生品,酒黄连则低于生品;而7种生物碱各自含量,因炮制方法不同而相异。(3)建立了黄连药材HPLC特征图谱测定方法,经方法学验证,该方法精密度、重现性、稳定性均符合要求;通过对15批黄连的特征图谱相似度的分析,找出共有特征峰15个,与黄连生物碱对照品比对后,指认了 8个峰,均为黄连生物碱成分。(4)采用四氧嘧啶小鼠糖尿病模型,对比了味连、雅连及其炮制品在降血糖方面的药效差异,结果表明,在降血糖方面各不同基源黄连与其不同炮制品效果差异不大,以萸雅连效果最为明显,在调节糖脂代谢紊乱和抗氧化方面,以雅连效果最佳。
[Abstract]:Rhizoma Coptidis is the dried rhizome of Coptischinensis Franch, Coptis deltoidea C.Y.Cheng et Hsiao. or Coptis teeta Wall. in autumn. The medicinal materials are divided into "Weilian", "Yalen" and "Yunlian". Coptis is a medicinal material produced in Sichuan Province, and Sichuan is the main region of production. In addition, a HPLC method for the simultaneous determination of alkaloids of 7 species of Coptis chinensis was established. The method was proved to be accurate, reproducible, precise and stable by using the traditional Sichuan variety Coptis omeiensis Coptis C.Y.Cheng.At first, the method was used to determine the alkaloids of 7 species of Coptis chinensis. The contents of 7 kinds of alkaloids and total alkaloids of 6 batches of Yalantii and 3 batches of Acanthopsis chinensis were determined, and the differences of total alkaloids and 7 kinds of alkaloids between different species were compared. The results showed that the contents of seven alkaloids were the highest among the three cultivars, and the contents of seven alkaloids were different from each other, and the differences were significant. 2) through the orthogonal experiments of four factors and three levels, the content of total alkaloids was the highest, and the content of seven alkaloids was different among the three varieties. Taking the total alkaloid content as the index, the optimum processing technology of Coptis chinensis, Rhizoma Corni and Coptis officinalis was screened. The contents of total alkaloids and seven kinds of alkaloids in different processed and raw products of Coptis chinensis were compared. The results showed that the content of total alkaloids of Cornel was the highest and higher than that of raw products, followed by Rhizoma Coptidis, and higher than that of raw ones. The content of 7 alkaloids, which were different from each other by different processing methods, was determined by HPLC characteristic chromatogram. The method was proved to be accurate, reproducible and stable. By analyzing the similarity of characteristic atlas of 15 batches of Coptis chinensis, 15 common characteristic peaks were found. After comparing with alkaloids of Coptis chinensis, 8 peaks were identified, all of which were alkaloid components of Coptis chinensis. The results showed that there was no significant difference in the effect of different basic sources of Rhizoma Coptidis and its processed products in lowering blood glucose, especially the effect of Corni's Artemisia officinalis was the most obvious, compared with its processed products, and the results showed that there was no significant difference in the effect of Rhizoma Coptidis and its processed products. Yalen has the best effect in regulating the disorder of glucose and lipid metabolism and anti-oxidation.
