汉族、维吾尔族乳腺癌患者与健康对照肠道菌群差异性研究

【摘要】:目的:1)采用IlluminaMiseq高通量测序技术,分别对维吾尔族和汉族乳腺癌患者与其对应的健康组妇女粪便样品进行肠道菌群的结构分析,研究维吾尔族和汉族乳腺癌患者各自肠道菌群结构及其多样性特征。探索不同民族肠道菌群与乳腺癌可能存在的相关性。初步构建不同民族乳腺癌肠道菌群构成情况,尝试筛选出与疾病显著相关的菌属。2)采用同种分析方法对汉族乳腺癌妇女和维吾尔族乳腺癌妇女肠道菌群进行差异性分析,初步探索两民族乳腺癌妇女肠道菌群在多样性及结构上的异同。方法:通过Illumina Miseq平台分别对汉族和维吾尔族乳腺癌妇女和其对应的健康妇女的16SrDNA V6-V8区进行高通量测序,测序结果进行OTU聚类分析、多样性及结构分析得出两个民族患者肠道菌群结构、丰度及多样性分布。结果:1)比较OTUs维吾尔族乳腺癌组(n=10)和健康对照组(n=10)的平均OTUs数目无显著差异(P0.05);汉族乳腺癌组(n = 25)的平均OTUs数目明显低于健康对照组(n=15)(P0.05)。利用稀释曲线、Chao1 指数、Shannon 指数和 Rank abundance 曲线等进行四组样本的复杂度分析,测序量足以覆盖肠道内各细菌种属,各组样品丰富度和均匀度较好。2)在属水平上,维吾尔族乳腺癌妇女与健康组存在显著性差异的菌属是拟杆菌属、毛螺菌科和杆菌属(P0.05);汉族乳腺癌妇女与健康组存在显著性差异的菌属是SubdoligranuLum菌属、Alistipes菌属、Blautia菌属、粪球菌属_3等在内的13个菌属(P0.05);维吾尔族乳腺癌组与汉族乳腺癌组中存在显著性差异的菌属是普氏菌属_9、拟杆菌属、Succinivibrio 菌属、SubdoligranuLum、Parabacteroides、Ruminococcus_2 等在内的19类菌属(P0.05);维吾尔族正常组与汉族正常组间存在显著性差异的菌属是拟杆菌属、Megamonas菌属、巨球型菌属、双歧杆菌属等在内的7个菌属(P0.05)。3)维吾尔族乳腺癌妇女组与健康妇女组、汉族乳腺癌妇女组与健康妇女组、汉族乳腺癌妇女组与维吾尔族乳腺癌妇女组、汉族健康妇女组与维吾尔族健康妇女组肠道菌群在各分类水平上均存在结构上的差异,且每一个组在每一个水平上均有其优势物种。结论:1)两民族乳腺癌患者与健康对照妇女肠道菌群之间存在着结构上的显著变化,提示肠道菌群结构上的这种变化和乳腺癌的疾病状态可能有某些关联。2)汉族乳腺癌妇女与维吾尔族乳腺癌妇女肠道菌群在结构组成上存在着差异,这种差异可能是遗传因素和环境因素共同作用的结果。3)健康汉族与健康组吾尔族妇女肠道菌群在结构比例上也存在着显著的差异,这种结果可能与基因型及饮食结构的不同有所关联。
[Abstract]:Objective: to analyze the intestinal flora of the fecal samples of women with breast cancer in Uygur and Han nationality and their corresponding healthy women by using IlluminaMiseq high-throughput sequencing technique.To study the structure and diversity of intestinal flora in Uygur and Han patients with breast cancer.To explore the possible correlation between intestinal microflora and breast cancer in different nationalities.The intestinal flora of breast cancer of Han nationality and Uygur nationality was analyzed by using the homologous analysis method, and the intestinal microflora of breast cancer of different nationalities was preliminarily constructed, and the bacteria related to the disease were screened out. 2) the difference of intestinal microflora between Han and Uygur breast cancer women was analyzed by the same method.To explore the diversity and structure of intestinal flora of breast cancer women.Methods: the 16SrDNA V6-V8 region of Han and Uygur breast cancer women and their corresponding healthy women were analyzed by high throughput sequencing using Illumina Miseq platform. The results were analyzed by OTU cluster analysis.The distribution of intestinal flora, abundance and diversity in patients with two nationalities were obtained by the analysis of diversity and structure.Results there was no significant difference in the average number of OTUs between the OTUs Uygur breast cancer group (n = 10) and the healthy control group (P 0.05), but the average OTUs number in the Han nationality breast cancer group (n = 25) was significantly lower than that in the healthy control group (P 0.05).The complexity analysis of four groups of samples was carried out by using the dilution curve Chao1 index Shannon index and Rank abundance curve. The sequencing amount was enough to cover the species and genera of bacteria in the intestinal tract, and the sample richness and evenness of each group was better than that of the other groups at the generic level.There were significant differences between the Uygur breast cancer women and the healthy group in the genera of Bacteroides, Helicobacteriaceae and Bacteriaceae (P0.05A), while the genera of SubdoligranuLum, Alistipes, Blautia, which were significantly different between the women of Han nationality and the healthy group, were found in this study.There are 13 strains of Staphylococcus spp. P0.05A, the genus of P0.05in Uygur breast cancer group and Han nationality breast cancer group, the genus P0.05in the normal Uygur group, and the subdoligranacteroides Parabacteroides Ruminococcus2 in the subdoligranacteroides Ruminococcus sp. 2 in the normal group of Uygur nationality, and the genus 9 in the genus Precinivibrio and the other 19 species in the genus Subdoligrania granuloca, Parabacteroides, Ruminococcus2, etc., respectively.The genus Megamonas, which has significant difference between the Han nationality and the normal group, is the genus Megamonas.The seven bacteria of the genus Bifidobacterium and megagonal type belong to P0.05. 3) Women with breast cancer of Uygur nationality and healthy women, women with breast cancer of Han nationality and healthy women, women with breast cancer of Han nationality and women with breast cancer of Uygur nationality.There were structural differences in intestinal flora between healthy women of Han nationality and Uygur nationality healthy women groups, and each group had its dominant species at each level.Conclusion (1) there were significant structural changes in intestinal flora between breast cancer patients and healthy women.It is suggested that there may be some correlation between the changes of intestinal flora structure and the disease status of breast cancer. 2) there are differences in the composition of intestinal microflora between women with breast cancer of Han nationality and women with breast cancer of Uygur nationality.This difference may be the result of the combination of genetic factors and environmental factors. 3) there are significant differences in the proportion of intestinal flora between healthy Han and healthy women of Wuer nationality.The results may be associated with different genotypes and dietary patterns.
