白虎加人参汤对糖尿病大鼠模型TRPA1通道表达影响的研究

【摘要】:目的:研究白虎加人参汤对糖尿病大鼠模型血清中丙二醛(MDA)及背根神经节(Drg)中TRPA1mRNA表达水平的影响,探讨白虎加人参汤对糖尿病及其并发症的疗效及其作用机制,为临床应用提供实验依据。方法:清洁级健康雄性SD大鼠45只,体重220±10g,普通饲料适应性喂养1周,随机选取10只作为空白对照组,剩余35只禁食12小时后给予单次腹腔注射链脲佐菌素(STZ)50mg/kg,72小时后尾静脉采血,共31只大鼠血糖≥16.7mmol/L,糖尿病大鼠模型造模成功,4只血糖未达标,造模不成功,成膜后大鼠随机分为白虎加人参汤组10只、α-硫辛酸组10只、模型对照组11只。白虎加人参汤组给予9.38g/kg/d灌胃,α-硫辛酸组给予20mg/kg/d腹腔注射,空白对照组、模型对照组给予同体积生理盐水灌胃,8周后测定相关指标,ELISA法测定大鼠血糖、血清MDA含量;光镜观察大鼠坐骨神经病理形态学改变;RT-PCR检测大鼠背根神经节中TRPA1mRNA的表达水平。结果:1、大鼠体重:治疗前各组大鼠体重基本相同,无统计学意义(P0.05);治疗后白虎加人参汤组、α-硫辛酸组大鼠体重较空白对照组体重增长缓慢,具有统计学意义(P0.01),较模型对照组体重增长快,具有统计学意义(P0.05),白虎加人参汤组和α-硫辛酸组比较,无统计学意义(P0.05)。2、大鼠血糖:造模72h后,与空白对照组比较,模型对照组、白虎加人参汤组和α-硫辛酸组大鼠血糖均升高,具有统计学意义(P0.01)。治疗后,白虎加人参汤组和α-硫辛酸组大鼠血糖较模型组下降,具有统计学意义(P0.01),白虎加人参汤组和α-硫辛酸组比较,白虎加人参汤组大鼠血糖下降较多,具有统计学意义(P0.05)。3、大鼠血清MDA:与空白对照组比较,模型对照组、白虎加人参汤组和α-硫辛酸组大鼠血清MDA水平升高,具有统计学意义(P0.05);与模型对照组比较,白虎加人参汤组和α-硫辛酸组大鼠血清MDA水平均下降,具有统计学差异(P0.01);白虎加人参汤组和α-硫辛酸组比较,无统计学意义(P0.05)。4、大鼠坐骨神经形态学光镜观察:空白对照组大鼠坐骨神经纤维排列整齐,结构清楚,神经束模连续完整,厚度基本一致,细胞核清晰,轴索、髓鞘结构完整;模型对照组神经纤维排列紊乱,分布不均,神经束模断裂破损,厚度不均匀,郎飞氏结间隙增宽,细胞核固缩,轴索变性,肿胀或皱缩,髓鞘呈节段性脱失;白虎加人参汤组和α-硫辛酸组神经纤维排列较整齐,束模部分断裂,部分细胞核固缩,部分轴索肿胀或皱缩,髓鞘个别脱失,程度均较模型对照组轻。5、大鼠背根神经节细胞TRPA1mRNA表达水平:与空白对照组比较,模型对照组、白虎加人参汤组和α-硫辛酸组大鼠背根神经节细胞中TRPA1mRNA的表达升高,具有统计学意义(P0.01);与模型对照组比较,白虎加人参汤组、硫辛酸组大鼠背根神经节细胞TRPA1mRNA的表达均下降,具有统计学意义(P0.05);白虎加人参汤组和α-硫辛酸组比较,白虎加人参汤组下降更明显,具有统计学差异(P0.05)。结论:1、白虎加人参汤可以降低糖尿病大鼠血糖。2、白虎加人参汤可以降低糖尿病大鼠血清MDA的含量减轻氧化应激状态。3、白虎加人参汤可以改善糖尿病大鼠坐骨神经病理形态。4、白虎加人参汤可以下调糖尿病大鼠背根神经节中TRPA1mRNA的表达。
[Abstract]:Objective: To study the effect of white tiger and human ginseng soup on the level of TRPA1mRNA expression of malondialdehyde (MDA) and dorsal root ganglion (Drg) in the serum of diabetic rats, and to explore the effect and mechanism of the white tiger plus human Ginseng Decoction on diabetes and its complications and to provide experimental basis for clinical application. Methods: 45 healthy male SD rats with a clean grade and 220 body weight were used. For 1 weeks, 10 rats were randomly selected as blank control group for 1 weeks. The remaining 35 rats were given a single intraperitoneal injection of streptozotocin (STZ) 50mg/kg after 12 hours of fasting, and after 72 hours the tail vein was collected. A total of 31 rats had blood sugar above 16.7mmol/L. The model of diabetic rats was successfully built, 4 blood glucose failed to reach the standard, and the model was unsuccessful. After the formation of the model, the membrane formation was unsuccessful and membrane formation after forming the membrane. Rats were randomly divided into 10 rats in the white tiger plus Shen Shen Decoction group, 10 in the alpha lipoic acid group and 11 in the model control group. The white tiger plus human ginseng decoction group was given 9.38g/kg/d gavage, the alpha lipoic acid group was given 20mg/kg/d intraperitoneal injection and the control group was given the control group. The model control group was given the same volume of normal saline, and the related indexes were measured after 8 weeks. The blood glucose of rats was measured by ELISA method and the serum M was measured. DA content; the pathological changes of the sciatic nerve in rats were observed by light microscope, and the expression level of TRPA1mRNA in the dorsal root ganglion of rats was detected by RT-PCR. Results: 1, the weight of rats in the rats before treatment was basically the same, without statistical significance (P0.05); the weight of the rats in the group of alpha lipoic acid group after treatment was higher than that of the blank control group. Slow, statistically significant (P0.01), compared with the model control group, the body weight increased rapidly and had statistical significance (P0.05). The white tiger plus Shen Shen Decoction group and the alpha lipoic acid group had no statistical significance (P0.05).2, the rat blood sugar was compared with the blank control group, the model control group, the white tiger addition Shen Shen Decoction group and the alpha lipoic acid group increased the blood sugar level. Higher, with statistical significance (P0.01). After treatment, the blood glucose of the White Tiger Group and the alpha lipoic acid group decreased compared with the model group, with statistical significance (P0.01). Compared with the white tiger and the alpha lipoic acid group and the alpha lipoic acid group, the blood glucose decreased more in the white tiger and the human ginseng decoction group, with statistical significance (P0.05).3, the rat serum MDA: and the blank control group Compared with the model control group, the serum MDA level of the rats in the white tiger plus human ginseng soup group and the alpha lipoic acid group increased, with statistical significance (P0.05). Compared with the model control group, the serum MDA level of the white tiger and the alpha lipoic acid group decreased with statistical difference (P0.01), and the white tiger plus human ginseng decoction group and the alpha lipoic acid group were not statistically significant. Study significance (P0.05).4, rat sciatic nerve morphological light microscope observation: the blank control group the sciatic nerve fibers arranged neatly, the structure is clear, the nerve bundle model is continuous and complete, the thickness is basically the same, the nucleus clear, the axon, the myelin sheath structure complete; the model control group the nerve fiber arrangement is disorderly, the distribution is uneven, the nerve bundle mode fracture breakage, thickness not Even, the clearance of the Lang flying junction widened, nucleus retraction, axonal degeneration, swelling or shrinkage, the myelin sheath showed segmental loss; the white tiger and the alpha lipoic acid group and the group of alpha lipoic acid group were neatly arranged, part of the nucleus was fractured, some nuclei were retracted, some axons swelled or crinkled, and the myelin sheath was individually lost, and the degree was less.5 than the model control group. The expression level of TRPA1mRNA in root ganglion cells: compared with the blank control group, the expression of TRPA1mRNA in the dorsal root ganglion cells of the model control group, the white tiger adding Shen Shen Decoction group and the alpha lipoic acid group increased, with statistical significance (P0.01). Compared with the model control group, the white tiger plus ginseng decoction group and the rat dorsal root ganglion cells of the lipoic acid group were TRPA1mRNA. The expression of the white tiger plus human ginseng decoction group and the group of alpha lipoic acid, white tiger and human ginseng soup group decreased more obviously, with statistical difference (P0.05). Conclusion: 1, white tiger plus Shen Shen decoction can reduce the blood sugar.2 of diabetic rats, white tiger plus ginseng decoction can reduce the content of serum MDA in diabetic rats and reduce oxidation. Stress state.3, white tiger plus Shen Shen decoction can improve the pathological morphological.4 of sciatic nerve in diabetic rats, and white tiger plus Shen Shen decoction can reduce the expression of TRPA1mRNA in the dorsal root ganglion of diabetic rats.
