pDNA-EDA和pDNA-EGF的协同作用对于汗腺再生影响

【摘要】:目的:在诱导汗腺再生的作用上,pDNA-EDA和pDNA-EGF的协同作用是否比单一的pDNA-EDA的作用更有效。方法:事先制备的负载骨髓间充质干细胞(MSC)的基因活性胶原-壳聚糖支架。以加入pDNA-EDA的负载MSC的胶原一壳聚糖/硅橡胶双层皮肤支架为对照组(EDA组),以加入pDNA-EGF和pDNA-EDA的负载MSC的胶原—壳聚糖/硅橡胶双层皮肤支架为实验组(EDA+EGF组)。建立SD大鼠脚掌的全层皮肤损伤模型,将以上两种支架植入皮肤缺损中。术后2周、4周、8周分别大体观察伤口愈合情况,并取再生组织标本做切片HE染色分析组织学观察,采用免疫组化的方法鉴定再生结构是否为汗腺组织。结果:术后2周、4周、8周大体观察对照组(EDA组)与实验组(EDA+EGF组)在伤口修复的进度上没有明显差别,第8周时两组大鼠脚掌皮肤均已均愈合完好,且无明显疤痕遗留。对2周、4周和8周的组织切片进行HE染色分析,发现在8周时实验组(EDA+EGF组)切片中的组织结构与正常类似,可见大量脂肪细胞,并出现类汗腺结构,对照组(EDA组)的切片中大部分细胞还是成纤维细胞,也浙江大学硕士学位论文 中文摘要发现类似汗腺的结构,但数量较少,结构较简单。经CK8和CK14鉴定阳性,表明该结构为汗腺。结论:以胶原一壳聚糖/硅橡胶双层皮肤支架为负载容器,pDNA-EDA+pDNA-EGF的协同作用诱导SD大鼠MSC向汗腺分化的能力比单独的pDNA-EDA强。
[Abstract]:Aim: whether the synergistic effect of pDNA-EDA and pDNA-EGF is more effective than that of single pDNA-EDA in inducing sweat gland regeneration. Methods: the gene active collagen-chitosan scaffold of MSCs loaded with bone marrow mesenchymal stem cells (MSCs) was prepared in advance. The collagen-chitosan / silicone rubber bilayer skin scaffold loaded with pDNA-EDA was used as the control group, and the collagen-chitosan / silicone rubber double-layer skin scaffold loaded with MSC with pDNA-EGF and pDNA-EDA was used as the experimental group. The full-thickness skin injury model of SD rats was established and the above two scaffolds were implanted into the skin defect. The wound healing was observed at 2 weeks, 4 weeks and 8 weeks after operation, and the tissue sections of regenerated tissue were analyzed by HE staining. Immunohistochemistry was used to identify whether the regenerated structure was sweat gland tissue. Results: there was no significant difference in the progress of wound repair between the control group and the experimental group at 2 weeks and 4 weeks and 8 weeks after operation. At the 8th week, the paw skin of the two groups had healed well, and there was no obvious scar left over. The histological structure of the tissue sections of the experimental group (EDA EGF group) was similar to that of the control group at the 8th week. A large number of adipocytes and sweat gland like structure were observed in the tissue sections at the 2nd week and 4th week and 8th week, respectively. In the control group (EDA group), most of the cells were still fibroblasts. The structure similar to sweat glands was found in Zhejiang University Master's degree thesis in Chinese, but the number was small and the structure was simple. The structure was identified as sweat gland by CK8 and CK14. Conclusion: the synergistic effect of collagen-chitosan / silicone rubber bilayer skin scaffold pDNA-EDA pDNA-EGF on inducing the differentiation of MSC into sweat glands in SD rats is stronger than that of pDNA-EDA alone.
