ADAM33基因V4位点多态性、维生素D水平与乌鲁木齐地区儿童支气管哮喘的相关性研究

【摘要】:目的:探讨乌鲁木齐地区支气管哮喘儿童ADAM33基因V4位点多态性、维生素D(VitD)水平及其相关性研究,期望从基因水平为指导喘患儿的防治提供理论依据。方法:选取同期5-15岁于乌鲁木齐地区生活的哮喘患儿154人(其中汉族111,维族43),同时设立对照组健康非哮喘儿童120人(汉族64,维族56),首先采用PCR聚合酶链反应对ADAM 33基因V4位点进行单核苷酸多态性(SNP)分型,并用ELISA法检测VitD水平,进一步根据民族进行组内分层,进行V4位点SNP、VitD分析并进行相关性分析。结果:(1)V4位点3种基因型频率分布在病例组和对照组间差异有统计学意义(x2=6.806,P=0.033),在不同民族间差异无统计学意义(实验组:x2=0.714,P=0.699;对照组:x2=5.898,P=0.052),考虑ADAM 33基因V4位点与乌鲁木齐地区维吾尔族、汉族儿童哮喘发病均不相关。(2)VitD水平检测病例组与对照组数值有差异,差异有统计学意义(t=2.341,P=0.020)。(3)ADAM 33基因V4位点与VitD水平相关性分析。结论:(1)ADAM33基因V4位点与乌鲁木齐地区支气管哮喘儿童发病有相关性,但无民族间差异。V4位点突变后的G基因可能是支气管哮喘发病的危险因素。(2)VitD水平与支气管哮喘儿童发病有关。(3)ADAM 33基因V4位点不影响VitD水平。
[Abstract]:Objective: to investigate the V4 polymorphism of ADAM33 gene and vitamin D Vitamin D level in children with bronchial asthma in Urumqi. Methods: 154 asthmatic children aged 5 to 15 years living in Urumqi were selected (111 Han, 43 Uygur) and 120 healthy non-asthmatic children (Han 64, Uygur 56) in the same period. PCR polymerase chain reaction (PCR) was used to detect Adam. Single nucleotide polymorphisms (SNPs) were performed at V4 locus of 33 gene. The level of Vit D was detected by Elisa, and further stratified according to ethnic group, the V4 SNPV VitD was analyzed and the correlation analysis was carried out. Results the frequencies of three genotypes at the V4 locus were significantly different between the case group and the control group, but there was no significant difference among the different nationalities (experimental group: x20.714, P0. 699; control group: x2 + 5. 898P0. 052), considering the V4 locus of ADAM33 gene and Uygur nationality in Urumqi region. The level of VitD in asthmatic children of Han nationality was not related to the incidence of asthma. The V4 locus of ADAM33 gene and the level of VitD were significantly different between the case group and the control group, and the difference was statistically significant between the V4 locus of ADAM33 gene and the level of VitD. Conclusion the V4 locus of ADAM33 gene is associated with the incidence of bronchial asthma in Urumqi. However, the G gene after mutation of V4 locus in different nationalities may be the risk factor of asthma. The V4 locus of ADAM33 gene has no effect on the level of VitD in children with bronchial asthma.
