【摘要】:目的:探讨在急性病毒性心肌炎小鼠发病过程中,是否存在骨髓来源CD45~+CD34~+VLA-4~+细胞动员及其意义。方法:1、应用流式细胞术检测CD45~+CD34~+VLA-4~+细胞在心肌、外周血和骨髓中的变化;2、免疫组化法检测心肌组织中定植因子VCAM-1的表达情况;3、实时荧光定量PCR法检测心肌组织中VCAM-1与炎症因子IL-1β、TNF-αmRNA的相对表达量;4、Western-blotting法检测定VCAM-1与IL-1β、TNF-α蛋白的相对表达量?结果:1、急性病毒性心肌炎小鼠心肌中CD45~+CD34~+VLA-4~+细胞的表达与对照组比较,第3天升高,第7天达到高峰,之后开始下降,第14天和第28天仍高于对照组(P0.05);外周血中及骨髓中CD45~+CD34~+VLA-4~+细胞的表达与对照组比较,第3天降低,但之后开始升高,第7天达到高峰,随后下降,第14天和第28天仍高于对照组(P0.05)。2、免疫组化示急性病毒性心肌炎小鼠心肌炎症细胞的周围VCAM-1的相对表达量明显增加,且伴多量的新生毛细血管生成。VCAM-1与心肌组织中的CD45~+CD34~+VLA-4~+细胞呈正相关关系。3、实时荧光定量PCR及Western-blotting示急性病毒性心肌炎小鼠心肌中VCAM-1及TNF-α、IL-1β的表达量明显高于对照组(P0.05)。VCAM-1与IL-1β、TNF-α的mRNA表达量呈正相关关系,VCAM-1的蛋白表达量与心肌组织中的CD45~+CD34~+VLA-4~+细胞呈正相关关系。4、动员到心肌组织中的CD45~+CD34~+VLA-4~+细胞与急性病毒性心肌炎心肌中的炎症因子及病理积分不成线性关系。结论:在病毒性心肌炎小鼠中,VCAM-1促进CD45~+CD34~+VLA-4~+细胞动员到心肌中,可能参与急性病毒性心肌炎的发病过程及心肌组织的再生与修复。
[Abstract]:Objective: To investigate whether there is mobilization and significance of bone marrow derived CD45~+CD34~+VLA-4~+ cells in the pathogenesis of acute viral myocarditis. Methods: 1, the changes of CD45~+CD34~+VLA-4~+ cells in the myocardium, peripheral blood and bone marrow were detected by flow cytometry; and 2, the expression of colonization factor VCAM-1 in myocardial tissue was detected by immunohistochemistry. 3, the relative expression of VCAM-1 and inflammatory factors IL-1 beta, TNF- alpha mRNA in myocardium was detected by real time fluorescence quantitative PCR. 4, Western-blotting method was used to detect the relative expression of VCAM-1 and IL-1 beta and TNF- alpha protein? Results: 1, the expression of CD45~+CD34~+VLA-4~+ cells in the myocardium of acute viral myocarditis was compared with the control group, third days up. High, seventh days reached the peak, then began to decline, fourteenth days and twenty-eighth days is still higher than the control group (P0.05), the expression of CD45~+CD34~+VLA-4~+ cells in the peripheral blood and bone marrow compared with the control group, third days lower, but then began to rise, the seventh day to reach the peak, then decreased, fourteenth days and twenty-eighth days is still higher than the control group (P0.05).2, immunohistochemistry showed urgent. The relative expression of VCAM-1 in the myocarditis and myoinflammatory cells of the mice with sexual viral myocarditis increased significantly, and the number of new capillary generated.VCAM-1 was positively related to the CD45~+CD34~+VLA-4~+ cells in the myocardium,.3, real-time fluorescent quantitative PCR and Western-blotting showed VCAM-1 and TNF- in the myocardium of acute viral myocarditis in mice The expression of alpha, IL-1 beta was significantly higher than that of the control group (P0.05).VCAM-1 and IL-1 beta, and the mRNA expression of TNF- a was positively correlated. The expression of VCAM-1 protein was positively correlated with CD45~+CD34~+VLA-4~+ cells in myocardial tissue. It was mobilized to the CD45~+CD34~+VLA-4~ + cells in the myocardium and the cause of inflammation in the myocardium of acute viral myocarditis. Conclusion: in the mice with viral myocarditis, VCAM-1 promotes the mobilization of CD45~+CD34~+VLA-4~+ cells into the myocardium, and may be involved in the pathogenesis of acute viral myocarditis and the regeneration and repair of myocardial tissue.
