三种乌蔹莓有效成分及华中乌蔹莓黄酮提取工艺和功能性研究

【摘要】:本文选取湖北省恩施州常见的葡萄科乌蔹莓属三种植物(乌蔹莓、三叶乌蔹莓、华中乌蔹莓)为试验材料,对其有效成分进行了相应研究,主要结论如下:1.三种乌蔹莓中黄酮含量最高,为主要有效成分,主要存在于叶片中;考虑生物量和有效成分含量,华中乌蔹莓为试验所讨论的三种乌蔹莓中最具开发意义的物种,黄酮为其最有开发意义的活性成分。2.对华中乌蔹莓醇提物进行成分分析,主要成分为:绿原酸、芦丁和槲皮素,芦丁占总醇提物百分比为40.41%,绿原酸为17.23%,槲皮素为7.14%,其中绿原酸在乌蔹莓属植物中首次发现,本文首次对华中乌蔹莓乙醇提取物进行了成分分析,芦丁为华中乌蔹莓主要黄酮类物质。3.经过单因素与正交试验优化,华中乌蔹莓黄酮最佳提取工艺为:料液比1:40,提取时间100 min,提取温度80℃,微波辅助时间3.0 min,在得出的最佳工艺条件下黄酮得率为131.93 mg/g。其中提取温度对黄酮得率影响最大,微波辅助时间对黄酮得率影响次之,料液比和提取时间对黄酮得率影响不显著。华中乌蔹莓黄酮的含量在全年中,6~10月含量较高,1~5月含量较低。4.华中乌蔹莓黄酮粗纯物能显著增强大鼠的抗氧化能力:短时间连续灌胃试验中,在0~150 mg/kg剂量范围内药量与药效呈现正相关趋势,随着剂量的增加,抗氧化效果越强,但达到150mg/kg后,随着剂量增加药效反而下降,短时间连续服用华中乌蔹莓黄酮时应注意剂量不能超过150 mg/kg;长期灌胃试验中在试验范围内,华中乌蔹莓黄酮粗纯物随着剂量的增加对大鼠有很好的抗氧化效果且药量与药效呈现正相关趋势,在一定的浓度上优于阳性对照VC。长期灌胃试验中,在试验范围内,低剂量和中剂量的华中乌蔹莓黄酮粗纯物对大鼠的抗疲劳有增效作用,高剂量组对其抗疲劳效果有显著性下降作用。
[Abstract]:In this paper, three plants of the genus Ampelopsis (Ampelopsis japonica, Ampelopsis japonica), which are common in Enshi Prefecture, Hubei Province, were selected as experimental materials, and their effective components were studied. The main conclusions are as follows: 1. The flavonoids content of the three kinds of Ampelopsis japonica is the highest, which is the main active component, and mainly exists in the leaves, considering the biomass and the content of the active components, the most exploitative species of the three kinds of Ampelopsis japonica discussed in the experiment is the Ampelopsis japonica. Flavonoids are the most important active components. The main components were Lv Yuan acid, rutin and quercetin. The percentage of rutin to total alcohol extract was 40.41. The ratio of Lv Yuan acid to total alcohol extract was 17.23, and quercetin was 7.14. Among them, Lv Yuan acid was found for the first time in the plants of the genus Ampelopsis, and the main components were: Lv Yuan acid, rutin and quercetin, the percentage of rutin to the total alcohol extract was 40.41. In this paper, the components of ethanol extracts from Ampelopsis japonica were analyzed for the first time. Rutin is the main flavonoids of Ampelopsis japonica. By single factor and orthogonal experiment, the optimum extraction technology of flavonoids from Ampelopsis japonica was as follows: ratio of material to liquid 1: 40, extraction time 100 min, extraction temperature 80 鈩,
