钠钙交换体亚型1介导CDX2上调促进Barrett食管发生发展的研究

目的:Barrett’s食管(Barrett’s Esophagus,BE)是发生于食管下段鳞柱状交界区的鳞状上皮被单层柱状上皮所替代的一种病理现象,常伴有肠上皮化生,是一种公认的食管腺癌的癌前病变。研究证实胆汁反流能够上调肠型分化标志物尾侧型同源盒转录因子-2(Caudal-related homeobox transcription factor-2,CDX2)的表达,从而促进BE的发生、发展,但具体机制不清。我们的前期研究发现脱氧胆酸(Deoxycholic acid,DCA)刺激正常食管上皮细胞过程中伴随着细胞内钙的增高,并促进正常食管上皮细胞中CDX2的表达。这一结果提示细胞内钙活化可能参与上调CDX2的表达。提示参与调控细胞内钙活化的离子通道在这一事件中起着举足轻重的作用。钙离子是细胞内重要的第二信使,参与了细胞多种生理与病理过程。钠钙交换体亚型1(Na+/Ca2+exchanger1,NCX1)是调控细胞内外钙平衡的重要离子通道之一。我们既往的研究发现,NCX1参与调控肝癌细胞内钙重构并促进肝癌细胞的增殖、分化。G蛋白偶联胆汁酸...
