基于transformer2和doublesex级联互作的烟粉虱性别决定机制研究

【摘要】:性别决定是生物两性形成和正常生长发育的基础,对种群延续意义重大。烟粉虱是一种世界性农业害虫,为单双倍体昆虫。雄虫来自未受精的卵为单倍体,雌虫来自受精卵为双倍体。为了研究烟粉虱的性别决定机制,对其性别决定中的两个关键基因transformer2(Bttra2)和doublesex(Btdsx)进行了克隆和分析,并进一步利用RNAi技术阐述了其在烟粉虱性别决定中的作用。研究结果表明烟粉虱Bttra2全长1702bp,由10个外显子和9个内含子组成;Btdsx全长2339bp,由7个外显子和6个内含子组成。发现Bttra2有四种剪接形式,编码四种蛋白,表达丰度雄虫高于雌虫;而Btdsx有13种剪接形式,编码六种蛋白,表达丰度雄虫也高于雌虫。明确了transformer2和Btdsx在烟粉虱性别决定通路中均发挥至关重要作用且两者相互影响和制约,Bttra2/Btdsx基因的干扰影响Vg基因和Btdsx/Bttra2的表达量,造成烟粉虱雄虫生殖器官尾针畸形。研究结果对于阐明烟粉虱性别决定机制具有重要理论意义。
[Abstract]:Sex determination is the basis for the formation of biological hermaphroditism and normal growth and development, which is of great significance for population continuation. Bemisia Tabaci is a worldwide agricultural pest, a haploidy insect. Males from unfertilized eggs were haploid and females from fertilized eggs were diploidy. In order to study the sex determination mechanism of Bemisia Tabaci, two key genes, transformer2Bttra2) and DoublesexexBtdsx), were cloned and analyzed, and their roles in sex determination of Bemisia Tabaci were further elucidated by using RNAi technique. The results showed that the total length of Bemisia Tabaci Bttra2 was 1702 BP, which consisted of 10 exons and 9 introns, which consisted of 7 exons and 6 introns. It was found that Bttra2 had four splicing forms, encoding four proteins, and that Btdsx had 13 splicing forms, encoding six proteins, and expressed more abundant male than female. It is clear that transformer2 and Btdsx play an important role in the sex-determining pathway of Bemisia Tabaci and that the interference of Bttra2 / Btdsx gene affects the expression of Vg gene and Btdsx/Bttra2 and causes malformation of male reproductive organ of Bemisia Tabaci. The results are of great theoretical significance to clarify the sex determination mechanism of Bemisia Tabaci (Bemisia Tabaci).
