槐属观赏乔木国槐、龙爪槐、金枝国槐和红花国槐抗旱性抗寒性生理研究

【摘要】:本文以国槐(Sophora japonica Linn.)、龙爪槐(Sophora japonica var.pendula Hort.)、金枝国槐(Sophora Japonica 'Golden Stem')和红花国槐(Sophora japonica 'Honghua')为试验材料,研究其抗旱抗寒生理特性。槐属植物是风景园林工作中常见的植物,北方地区夏季干旱冬季严寒,研究此4种植物的抗旱抗寒特性不仅对其在寒旱区的应用具有重要意义,还可为抗旱性和抗寒性分子机制研究提供数据支持。抗旱性试验选择聚乙二醇-6000(PEG-6000)模拟干旱胁迫处理方法;抗寒性试验选择人工低温胁迫和自然降温两种处理方法。研究内容如下:1.抗旱性研究对叶片相对含水量、相对电导率、叶绿素含量、叶绿素a/b值、丙二醛(MDA)含量等5个生理指标进行测定。其中相对含水量、叶绿素含量与抗旱性呈正相关;相对电导率、叶绿素a/b值、丙二醛含量与抗旱性呈负相关。方差分析可知,各处理间差异达到显著程度以上。运用隶属函数法进行综合评价,抗旱性由强到弱为:国槐红花国槐龙爪槐金枝国槐。2.人工低温胁迫试验选择相对电导率、可溶性糖含量、丙二醛含量等3个生理指标,相对电导率、丙二醛含量与植物抗寒性呈负相关,可溶性糖含量与植物抗寒性呈正相关。拟合Logistic方程得出国槐、红花国槐、金枝国槐、龙爪槐的半致死温度LT50分别为-24.89℃、-25.37℃、-21.87℃、-20.10℃,低于半致死温度的低温对植物产生不可恢复的伤害甚至导致死亡。3.自然降温胁迫分别测定4种植物一、二、三年生枝条的相对电导率的变化情况。生长时间越长的枝条,木质化程度越高,抗寒性越强。0℃以上的冷害对4种植物枝条的影响不显著,0℃以下的冻害对其影响显著。自然降温和人工低温胁迫对其细胞膜相对透性的影响基本一致,印证了人工低温胁迫试验结果的可靠性。运用隶属函数法进行综合评价,抗寒性由强到弱为:国槐红花国槐金枝国槐龙爪槐。4.对国槐等4种植物的抗旱性和抗寒性隶属函数值进行了聚类分析,结果显示4种植物中,国槐和红花国槐的抗逆性较好,且比较接近,适宜在园林工作中推广种植;金枝国槐的抗旱性最差,春夏易发生干旱的季节需进行适当浇灌;龙爪槐的抗寒性最差,在接近或低于其半致死温度的地区越冬需做保温防寒处理。
[Abstract]:The physiological characteristics of drought resistance and cold resistance of Robinia (Sophora japonica Linn.), (Sophora japonica var.pendula Hort.), (Sophora Japonica 'Golden Stem' and (Sophora japonica' Honghua' were studied in this paper. Sophora is a common plant in landscape architecture. It is of great significance to study the drought resistance and cold resistance characteristics of the four species in summer and winter in northern China, and it is of great significance to its application in cold and arid areas. It can also provide data support for the study of molecular mechanism of drought resistance and cold resistance. Polyethylene glycol-6000 (PEG-6000) was used to simulate drought stress in drought resistance test and artificial low temperature stress and natural cooling were used in cold resistance test. The contents of the study are as follows: 1. Five physiological indexes, such as relative water content, relative conductivity, chlorophyll content, chlorophyll a / b value and malondialdehyde (MDA) content, were measured in drought resistance study. The relative water content and chlorophyll content were positively correlated with drought resistance, while the relative conductivity, chlorophyll a / b value and malondialdehyde content were negatively correlated with drought resistance. Analysis of variance showed that the difference among the treatments was more than significant. By using the method of membership function, the drought resistance was evaluated from strong to weak: safflower, locust, dragon, claw, gold, branch and locust. Three physiological indexes, such as relative conductivity, soluble sugar content and malondialdehyde content, were selected in the artificial low temperature stress test. The relative conductivity and malondialdehyde content were negatively correlated with the cold resistance of plants, and the soluble sugar content was positively correlated with the cold resistance of plants.
