大白菜春化种子的蛋白质组分析

【摘要】:作为一种重要的成花诱导途径,春化作用在大多数冬性或者二年生植物实现营养生长向生殖生长的转变过程中起着不可替代的作用。大白菜属于典型的种子春化型植物,在种子萌发期便可感受低温处理,进而提前开花。育种上常利用春化作用来加快大白菜的培育进程,但目前大白菜种子春化的内在调控机理并不明了。因此,本研究运用iTRAQ技术研究了大白菜萌发种子在春化过程中的蛋白表达情况,以期为揭示大白菜种子春化的调控机制提供一定依据。主要研究内容和结果如下:1.对大白菜双单倍体'FT'萌发种子进行不同时间春化处理,随着春化天数的增加,大白菜所需抽薹开花的时间会变短,低温处理25d可有效促进'FT'度过春化。2.利用iTRAQ技术对大白菜25℃萌发24h种子(Ger),及该种子经4℃春化25d的种芽(Ver)和25℃生长60h的种芽(Nor,与Ver种芽等长)进行蛋白质组分析,共鉴定到4197个蛋白质,其中在Nor-VS-Ver、Ger-VS-Ver、Ger-VS-Nor中分别鉴定到641、604、644个差异表达蛋白。3.对Nor-VS-Ver中差异表达蛋白进行GO和KEGG功能富集分析。结果表明,发生显著变化的蛋白质主要来源于细胞质和细胞膜且作为一些蛋白酶或者分子伴侣等在大白菜体内的碳水化合物代谢,蛋白质代谢、次生物质合成以及逆境响应等生物过程中起重要作用。4.对Ger-VS-Ver和Ger-VS-Nor中的差异表达蛋白进行Venn图谱分析,筛选到329个Ger-VS-Ver特有差异表达蛋白,同样进行功能富集分析。结果表明,这些蛋白也涉及糖类、蛋白质、氨基酸和脂类等物质代谢,并且伴随能量代谢和信号转导等过程。5.采用Western blot技术检测2个GDSL脂肪酶家族蛋白(显著富集到类胡萝卜素代谢通路)的表达情况,qRT-PCR技术检测12个差异蛋白对应的基因(植物响应逆境、生长发育、抗氧化等过程相关)的表达情况,从蛋白水平和转录水平上验证了蛋白质组定量分析的准确性。6.对相同样品的蛋白质组和已有转录组进行联合分析,结果表明,差异表达蛋白与差异表达基因之间只有少数基因被关联到且相关性较低,也暗示着大白菜种子春化过程中蛋白质的表达存在转录后调控作用。
[Abstract]:As an important pathway of flower induction, vernalization plays an irreplaceable role in the transformation of vegetative growth to reproductive growth in most winter or biennial plants. Chinese cabbage (Brassica campestris L.) is a typical spring-seed plant, which can be treated with low temperature during seed germination, and then blossoms ahead of time. In breeding, vernalization is often used to speed up the cultivation of Chinese cabbage, but the internal regulation mechanism of Chinese cabbage seed vernalization is not clear at present. Therefore, iTRAQ technique was used to study the protein expression of germinating seeds of Chinese cabbage in the process of vernalization, in order to provide some basis for revealing the regulation mechanism of vernalization of Chinese cabbage seeds. The main contents and results of the study are as follows: 1. The time of bolting and flowering in Chinese cabbage (Brassica campestris L.) was shortened with the increase of the days of vernalization, and 25 days of low temperature treatment could effectively promote the 'FT' through vernalization. 2.
