中华绣线梅组织培养技术研究

【摘要】:中华绣线梅(Neillia sinensis Oliv.)为我国特有树种,主要分布在陕西、云南等地,属蔷薇科绣线菊亚科绣线梅属植物,是一种较具发展前景的野生花灌木。其树态清秀、叶形美丽、花团淡粉色、果实红色,集观花、观叶、观果为一体。花期5-6月,果期8-9月。叶片内含有山梨醇及鞣质,药用价值较高,因此在园林观赏价值方面及医学药用方面较具开发前景。本研究通过对中华绣线梅进行组织培养,以期最终获得一定数量的苗木,对其植物种质资源加以保护和利用,增加我国城市园林绿化树种的种类,丰富城市景观多样性,提高城市园林品位。本研究以中华绣线梅的腋芽茎段为外植体,探讨了初代培养阶段外植体消毒方式对其培养的影响;增殖培养阶段中最适宜组培苗扩繁增殖的植物生长调节剂浓度的配比方案;生根培养阶段中最适宜其生根的培养基类型及不同的激素种类和浓度对其生根的影响;炼苗移栽过程中应注意的事项。从外植体消毒建立无菌体系到增殖培养再到生根诱导及炼苗移栽等一系列的研究,为中华绣线梅组织培养技术提供了系统的依据。结果如下:1.中华绣线梅腋芽茎段的最佳消毒处理为75%酒精30s+0.1%HgCl2消毒8min。2.组培苗的最佳增殖处理为MS+6-BA 1.0mg/L+NAA 0.2mg/L组合,植株健壮、丛生芽多、长势好、芽颜色浓绿,生长迅速,其增殖系数最高为4.13,丛生芽高度为2.97cm。3.组培苗诱导生根的最佳基本培养基类型为1/4MS,最佳激素种类及浓度为IBA0.7mg/L,平均生根条数为5.03条、平均根长为2.58cm、生根率为63.3%。植株健壮、主根粗、须根多、叶片浓绿。4.选取植株生长健壮,高度在2cm以上的组培苗进行炼苗移栽,选择以蛭石:沙子:腐殖质(1:1:2)混合的基质作为炼苗培养的栽培基质,常规培养20d后,其成活率为36%。
[Abstract]:Chinese embroidered plum (Neillia sinensis Oliv.) It is a endemic tree species in China, mainly distributed in Shaanxi, Yunnan and other places. It is a kind of wild flower shrub, which belongs to the family of Rosaceae, Hydrangea subfamily, Hydrangea, and is a kind of wild flower shrub. Its tree-shaped, beautiful leaves, pale pink flowers, red fruit, set to see flowers, look at leaves, see fruit as a whole. Flowering period is 5-6 months, fruit stage is 8-9 months. The leaves contain sorbitol and tannin, which have high medicinal value, so it has more prospect in ornamental value and medical medicine. In this study, the tissue culture of Chinese embroidered plum was carried out in order to obtain a certain number of seedlings, protect and utilize its plant germplasm resources, increase the species of urban landscaping trees and enrich the diversity of urban landscape in China. Improve the taste of urban gardens. In this study, the axillary bud stem segment of Hydrangea chinensis was used as explants, and the effects of disinfection methods of explants on the culture were discussed, and the optimal concentration of plant growth regulators for the propagation and propagation of plantlets in the stage of proliferation and culture was also discussed in this paper, and the optimal concentration of plant growth regulators for the propagation and propagation of plantlets in tissue culture was studied. In the stage of rooting culture, the most suitable medium type for rooting and the effects of different hormone types and concentrations on rooting were discussed, and the matters needing attention in the process of transplanting seedlings were also discussed. A series of studies on the establishment of aseptic system by sterilizing explants, multiplication culture, rooting induction and plantlet transplanting were carried out, which provided a systematic basis for tissue culture technology of Hydrangea chinensis. The results are as follows: 1. The best disinfection treatment of axillary bud stem segment of Chinese embroidered plum was 75% alcohol 30s 0.1%HgCl2 disinfection for 8 min. 2. The best multiplication treatment of tissue cultured seedlings was MS 6-BA 1.0mg/L NAA 0.2mg/L combination. The plant was robust, had more buds, grew well, the bud color was thick green and grew rapidly, and its proliferation coefficient was up to 4.13, The height of tufted buds is 2.97 cm. 3. The best basic medium type for inducing rooting of plantlets in tissue culture was 1 ~ 4 MS, the average rooting number of IBA0.7mg/L, was 5.03, the average length of root was 2.58 cm, the rooting rate was 63.3%, and the optimum hormone type and concentration were 5.03, 2.58 cm and 63.3%, respectively. The plant is strong, the main root is thick, the beard root is many, the leaf blade is thick green. 4. The plantlets with robust growth and height above 2cm were selected to transplant the plantlets, and the medium mixed with vermiculite, sand and humus (1:1:2) was selected as the culture medium. After 20 days of conventional culture, the survival rate was 36%.
