【摘要】:目的建立人子宫内膜癌裸鼠皮下移植瘤模型。方法培养人子宫内膜癌细胞系Ishikawa,制成肿瘤细胞悬液,于裸鼠(10只)右侧肩胛背部皮下注射,每只裸鼠注射约1×107个细胞。接种后每日观察裸鼠精神、活动、饮食、排便等状况及是否有肿瘤生长、肿瘤生长速度等。以接种部位可见或触及肿瘤小结节为成瘤标准,若第一次移植失败则考虑行二次瘤细胞接种。发现肿瘤生长后每3 d计算肿瘤体积。当肿瘤最长径达15 mm或出现皮肤破溃时,立即处死荷瘤裸鼠;否则于接种第49天将荷瘤裸鼠处死、剥出肿瘤,测量瘤体长径,称取瘤体质量。取肿瘤组织切片后行HE染色,观察肿瘤组织病理变化。结果有1只裸鼠第一次接种失败,在第二次接种后移植成功,一次成瘤率为9/10。在接种第40天和第46天各发现1只裸鼠瘤体表面皮肤破溃,发现后脱颈处死荷瘤裸鼠,测得肿瘤最长径分别为12.2、12.6 mm,肿瘤体积分别为505.14、510.30 mm3,肿瘤质量分别为0.88、0.86 g;至接种第49天,存活的7只裸鼠移植瘤瘤体表面皮肤完整,无破溃缺损,处死裸鼠后完整剥离肿瘤,测得肿瘤最长径为11.7~14.0 mm,瘤重0.86~1.23 g。取移植瘤组织进行病理检查,肿瘤组织大体表现与镜下表现符合人子宫内膜癌的特点。结论采用将Ishikawa细胞株接种于裸鼠右侧肩胛皮下的方式成功建立人子宫内膜癌裸鼠皮下移植瘤模型,且移植瘤组织和细胞保留了人子宫内膜癌的病理特征。
[Abstract]:Objective to establish a model of subcutaneous human endometrial carcinoma in nude mice. Methods cultured human endometrial carcinoma cell line Ishikawa into tumor cell suspension in nude mice (10) right scapular subcutaneous injection, each mouse injection of approximately 1 x 107 cells. After inoculation of mice were observed daily spirit, activity, diet, defecation other conditions and whether there is tumor growth, tumor growth rate. The inoculation site visible or palpable tumors into tumor nodules, if the first graft failure is considered for the two time of tumor cell inoculation. Found tumor growth every 3 d after the tumor volume was calculated. When the tumor diameter reached 15 mm or broken skin break when the nude mice were sacrificed immediately; otherwise the inoculation in nude mice were sacrificed forty-ninth days, stripping of tumor, measuring tumor size, weighing tumor weight. The tumor tissue sections were stained by HE staining to observe the pathological changes of tumor tissue. The results of 1 mice The first vaccination failure in second after inoculation of transplant success, a tumor was found 1 9/10. nude mice tumor surface skin ulceration after inoculation for fortieth days and forty-sixth days after the discovery of sacrificed mice, measured the tumor diameter were 12.2,12.6 mm, tumor volume integral don't 505.14510.30 mm3, tumor weights were 0.88,0.86 g; to forty-ninth days of inoculation, tumor of 7 nude mice body surface skin survived intact, no ulceration defect, the nude mice were killed after the tumor was removed, measured the tumor diameter of 11.7~14.0 mm, 0.86~1.23 g. and tumor weight of tumor tissue for pathological examination of tumor tissue. Gross appearance and endoscopic features in line with the characteristics of human endometrial carcinoma. Conclusion the Ishikawa cell line was inoculated subcutaneously in nude mice the right scapular model was successfully established subcutaneous human endometrial carcinoma in nude mice, and the transplanted tumor tissues and cells. The pathological features of endometrial carcinoma were retained.
