脂联素对高糖环境下人心肌细胞增殖和凋亡的干预作用

【摘要】:目的:研究在高糖环境下及加入脂联素(Adiponectin, ADPN)后对人心肌细胞增殖、凋亡情况及氧化应激指标ROS的影响,揭示脂联素在糖尿病心肌病中保护心肌细胞的作用。 方法:将培养的人心肌细胞分为:正常对照组(N组)、高糖组(G组)、高糖+脂联素组(A组),分别培养24h,48h及72h,用MTT法(四甲基偶氮唑蓝)检测细胞增殖,采用Western blotting法检测各组细胞凋亡相关蛋白Bax, Bcl-2的表达,用荧光探针DCFH-DA检测心肌细胞内活性氧(ROS)水平,观察细胞凋亡。 结果::①MTT法检测人心肌细胞增殖情况,24h高糖组与高糖+脂联素组比较差异无统计学意义,48h和72h高糖组与对照组相比增殖明显减弱(p0.05),并且高糖+脂联素组细胞增殖较高糖组明显增加,差异有统计学意义(p0.05)。②高糖组较对照组中人心肌细胞bcl-2蛋白含量下降,Bax蛋白含量上升,加入脂联素后,bcl-2蛋白含量显著上升,Bax蛋白含量显著下降,差异有统计学意义(p0.05)。③ROS在高糖组表达较正常组升高,脂联素干预后,ROS表达水平下降接近对照组,差异有统计学意义(p0.05)。 结论:高糖的持续作用可以抑制人心肌细胞的增殖,增加人心肌细胞的氧化应激损伤;加入脂联素干预后,细胞的增殖活性增加,减少了对人心肌细胞的氧化应激损伤,能保护高糖环境下人心肌细胞。
[Abstract]:Objective: To study the effects of Adiponectin (ADPN) on the proliferation, apoptosis and oxidative stress index ROS of human cardiac myocytes, and to reveal the role of adiponectin in protecting cardiomyocytes from diabetic cardiomyopathy.
Methods: the cultured myocardial cells were divided into normal control group (N group), high glucose group (G group), high glucose + adiponectin group (A group), 48h and 72h were cultured in 24h, using the MTT method (four methyl thiazolyl tetrazolium) cell proliferation assay, using Western blotting method to detect the fine apoptosis related protein Bax, Bcl-2 expression in myocardial cells of DCFH-DA fluorescent probe for detection of reactive oxygen species (ROS) level, the cell apoptosis was observed.
Results: the detection of human myocardial cell proliferation MTT assay, 24h high glucose group and high glucose + adiponectin group had no significant difference, 48h and 72h in high glucose group compared with the control group, the proliferation was significantly decreased (P0.05), and the proliferation of high glucose + adiponectin group were significantly increased compared with the high glucose group, the difference was statistically significant (P0.05). The high glucose group was lower than that of control group in the myocardial cell Bcl-2 protein, Bax protein content increased, adding adiponectin, significantly increased the content of Bcl-2 protein, Bax protein content decreased significantly, the difference was statistically significant (P0.05). The expression of ROS in high glucose group than in the normal group, adiponectin intervention, the expression level of ROS decreased to the control group, the difference was statistically significant (P0.05).
Conclusion: the effect of high glucose can inhibit myocardial cell proliferation, increase the damage of oxidative stress; adding adiponectin intervention, cell proliferation activity increased, reducing the oxidative stress injury of myocardial cells and protects human myocardial cells in high glucose environment.
