神经营养因子NT4及其受体在大鼠肺纤维化中的表达

【摘要】:实验目的 本研究通过对大鼠实验性肺纤维化中NT4及其受体TrkB的检测,来探讨NT4及TrkB与大鼠肺纤维化发生发展的关系,从而研究神经营养因子在肺纤维化中的变化,为肺纤维化发病机制的研究开辟新的途径。 实验方法 选wistar健康雄性大鼠30只作为研究的对象,分为对照组(向气管内注射生理盐水)、模型组(向气管内灌注博来霉素)两组;两组大鼠分别于造模后第7天、第14天、第28天各处死5只,取出大鼠肺组织,用Masson染色及HE染色来比较造模程度,用PCR和免疫组化检测模型组及对照组中NT4及TrkB的含量。应用SPSS17.0统计软件进行数据统计分析,单个因素的各时间点之间的比较,采用单因素方差分析,两两比较用t检验,并进行相关性分析。 实验结果 模型组造模后第7天、第14天、第28天NT4及TrkB均有不同程度的增高,且均随病变严重程度的增高而增高,结果符合统计学意义(P0.05),二者呈正相关(P0.001);对照组在对应时间段二者无明显变化(P0.05)。 实验结论 神经营养因子NT4及其受体TrkB在肺纤维化大鼠中表达增加,可能与肺上皮细胞增生及TrkB/NT4(?)由的信号传导被扰乱等因素有关,说明神经营养因子NT4及其受体参与肺纤维化的发生发展过程,为肺纤维化的发生提供新的论点。
[Abstract]:Experimental purpose The purpose of this study was to investigate the relationship between NT4 and TrkB and the occurrence and development of pulmonary fibrosis in rats by the detection of NT4 and its receptor TrkB in experimental pulmonary fibrosis. Therefore, to study the changes of neurotrophic factors in pulmonary fibrosis, and to open up a new way for the study of the pathogenesis of pulmonary fibrosis. Experimental method Thirty male wistar rats were divided into two groups: control group (intratracheal injection of normal saline) and model group (intratracheal instillation of bleomycin). Five rats were killed in each group on the 7th, 14th and 28th day after modeling. The lung tissue was taken out, and the degree of the model was compared by Masson staining and HE staining. The contents of NT4 and TrkB in model group and control group were detected by PCR and immunohistochemistry. The statistical data were analyzed by SPSS17.0 software and the time points of single factor were compared. Single factor analysis of variance (ANOVA) was used, and t test was used for pairwise comparison, and correlation analysis was carried out. Experimental results In the model group, NT4 and TrkB increased in varying degrees on the 7th, 14th and 28th day after the establishment of the model, and both increased with the severity of the lesion. The results were in accordance with the statistical significance (P 0.05), and there was a positive correlation between them. In the control group, there was no significant change in the two groups in the corresponding time period (P 0.05). Experimental conclusion The expression of neurotrophic factor NT4 and its receptor TrkB is increased in pulmonary fibrosis rats, which may be related to the proliferation of pulmonary epithelial cells and TrkB / NT4? The signal transduction was disturbed, which indicated that neurotrophic factor NT4 and its receptor were involved in the development of pulmonary fibrosis, and provided a new argument for the occurrence of pulmonary fibrosis.
