通络救脑口服液对快速老化模型小鼠RAGE和Aβ的影响

【摘要】:目的探讨通络救脑口服液对快速老化模型(SAMP8)小鼠晚期糖基化终末产物受体(RAGE)和淀粉样蛋白β(Aβ)表达的影响。方法 24只SAMP8小鼠随机分为模型组、通络救脑高、低剂量组(9,6 m L·kg~(-1)·d~(-1)),另设抗快速衰老亚系1(SAMR1)小鼠对照组(正常对照组),连续灌胃给药21 d。取海马区脑组织,分别采用酶联免疫吸附法(ELISA)和免疫组织化学法检测小鼠大脑海马内Aβ以及RAGE的表达。结果与模型组比较,通络救脑高、低剂量组海马区脑组织RAGE和Aβ表达降低(P0.01),RAGE蛋白含量明显降低(P0.01)。结论通络救脑口服液可通过抑制海马区脑组织RAGE和Aβ的表达,以减轻RAGE和Aβ介导的神经病理损伤作用。
[Abstract]:Objective to investigate the effect of Tongluojionao oral liquid on the expression of advanced glycosylation end product receptor (RAGEG) and amyloid 尾 -butadiene (A 尾) in mice with rapid aging model (SAMP8). Methods Twenty-four SAMP8 mice were randomly divided into two groups: model group, Tongluo Jiao Gao group, low dose group, 9m L / kg ~ (-1) D ~ (-1) ~ (-1) D ~ (-1), and control group (normal control group, n = 21) with anti-aging subline 1 ~ (1) SAM ~ (1) (control group), which were given intragastric administration continuously for 21 days. The expressions of A 尾 and RAGE in hippocampus of mice were detected by Elisa and immunohistochemistry. Results compared with the model group, the expression of RAGE and A 尾 in the hippocampus of the low dose group was significantly lower than that in the model group. Conclusion Tongluojionao Oral liquid can attenuate the neuropathological injury mediated by RAGE and A 尾 by inhibiting the expression of RAGE and A 尾 in hippocampus.
