VX-2组织悬液原位种植与Panc-1细胞悬液原位种植在家兔胰腺癌模型建立中的效果比较

【摘要】:目的目前有关家兔的胰腺癌模型制作方法报道较为少见,文中比较Panc-1细胞悬液原位种植与VX-2组织悬液原位种植在家兔胰腺癌模型建立中的效果。方法选取健康家兔30只,采取随机数字表法分为组织悬液组(n=15)、细胞悬液组(n=15)。组织悬液组采取VX-2组织悬液原位种植,细胞悬液组采取Panc-1细胞悬液原位种植,种植后观察模型的建立情况。通过B超、3.0T磁共振以及CT等辅助检查评价两种建模结果。结果组织悬液组第3周内有5只家兔出现十二指肠、结肠、盲肠、腹膜壁出现大量肿瘤组织种植性转移,3只家兔大网膜出现肿瘤组织种植性转移,MR T2见胃体后高信号影。细胞悬液组第3周内1只家兔十二指肠出现肿瘤组织种植性转移,MR LAVA见胃体后方稍高信号影。组织悬液组中15只模型全部建立成功,细胞悬液组仅发现3例建模成功。与组织悬液组比较,细胞悬液组第3、4周肿瘤种植成功率明显降低(46.66%vs 6.67%,100%vs 20.00%,P0.05)。结论 VX-2组织悬液原位种植较Panc-1细胞悬液原位种植的更具有可行性,更易实施。
[Abstract]:Objective to compare the effect of in situ implantation of Panc-1 cell suspension and VX-2 tissue suspension on the establishment of rabbit pancreatic carcinoma model. Methods 30 healthy rabbits were randomly divided into tissue suspension group (n = 15) and cell suspension group (n = 15). VX-2 tissue suspension was implanted in situ in the tissue suspension group and Panc-1 cell suspension was implanted in the cell suspension group. The establishment of the model was observed after implantation. The modeling results were evaluated by 3. 0T magnetic resonance imaging (MRI) and computed tomography (CT). Results in the third week of tissue suspension group, there were 5 rabbits with duodenum, colon, cecum and peritoneal wall with a large number of implanted metastasis. 3 rabbits had tumor tissue implantation metastasis in the greater omentum. Mr T2 showed high signal intensity behind the gastric body. In the third week of the cell suspension group, a rabbit duodenum showed implanted metastasis of tumor tissue. Mr LAVA showed a little hyperintense image behind the gastric body. In the tissue suspension group, all the 15 models were successfully established, and only 3 cases were successfully established in the cell suspension group. Compared with the tissue suspension group, the success rate of tumor implantation in the cell suspension group was significantly lower than that in the cell suspension group at the 3rd week (46.66%vs 6.67 vs 20.00% P0.05). Conclusion in situ implantation of VX-2 tissue suspension is more feasible and easier than that of Panc-1 cell suspension in situ implantation.
