人卵巢癌HO-8910原位移植瘤动物模型建立及分期研究

【摘要】:目的建立HO-8910卵巢癌荧光原位移植瘤模型,并进行分期研究。方法将人卵巢癌HO-8910细胞株在细胞培养瓶中培养至10代,选荧光稳定表达的细胞注射到裸鼠皮下,待成瘤后通过显微外科手术将体积为1mm3肿瘤块植入到实验裸鼠卵巢黏膜上,在活体成像技术下观察肿瘤的生长和转移情况,并应用病理学检测方法对肿瘤组织及附近组织进行观察。结果活体成像技术成功追踪了肿瘤的发展与转移,病理结果显示肿瘤细胞的转移趋势,2者与中医临床卵巢癌分期相结合,确立了卵巢癌分期。结论成功建立卵巢癌原位移植瘤模型,再现了肿瘤临床演变过程。其3期分期为肿瘤研究及抗肿瘤药物疗效判定研究提供了实验基础。
[Abstract]:Objective to establish a fluorescent orthopaedic transplantation model of HO-8910 ovarian cancer and study its staging. Methods Human ovarian cancer cell line HO-8910 was cultured in cell culture bottle for 10 generations. The cells stably expressed by fluorescence were injected into the skin of nude mice. After the tumor was formed, the volume of 1mm3 tumor was implanted into the ovarian mucosa of experimental nude mice by microsurgery. The growth and metastasis of the tumor were observed by in vivo imaging technique, and the tumor tissue and nearby tissues were observed by pathological examination. Results the development and metastasis of tumor were successfully followed by in vivo imaging. The pathological results showed the metastasis trend of tumor cells. The two were combined with the clinical stage of ovarian cancer in traditional Chinese medicine to establish the stage of ovarian cancer. Conclusion the orthotopic transplantation model of ovarian cancer was successfully established and the clinical evolution of the tumor was reproduced. The stage 3 stage provides an experimental basis for tumor research and antineoplastic drug efficacy evaluation.
