【摘要】:目的 探讨培养液中添加人成熟卵泡液(HFF)、表皮生长因子(EGF)对人未成熟卵母细胞体外成熟(IVM)、体外受精(IVF)及其胚胎发育的影响。 方法 未成熟卵母细胞来自接受IVM治疗的多囊卵巢综合症(PCOS)患者。病人随机分配到不同组别中:对照组:TCM199+10%SSS+0.075IU/ml FSH+0.51U/mlHCG+1μg/ml 17β-E_2+0.05mg/ml青霉素+0.075mg/ml链霉素+0.2mM/L丙酮酸;卵泡液组:对照组液+10%HFF:EGF组:对照组液+2ng/mlEGF。培养24-48小时后,对已排出第一极体的卵母细胞行卵胞浆内单精子显微注射(ICSI),注射后16-18h小时,观察卵母细胞受精情况,受精后继续培养2—3天行胚胎移植(ET),观察成熟率、受精率、卵裂率、2细胞、8细胞、优质胚胎率及种植率、临床妊娠情况。 结果 1、HFF、EGF对未成熟卵母细胞MII、受精及卵裂率的影响 HFF组与对照组相比,前者的卵裂率明显高于后者,P0.01,而MII率、受精率差异无显著性,P
[Abstract]:Objective to investigate the effect of human mature follicular fluid (HFF) and epidermal growth factor (EGF) on immature oocyte maturation (IVM), in vitro fertilization (IVF) and embryo development in vitro.
Methods immature oocytes from IVM treated with polycystic ovary syndrome (PCOS) patients. The patients were randomly assigned to different groups: the control group: TCM199+10%SSS+0.075IU/ml FSH+0.51U/mlHCG+1 g/ml -E_2+0.05mg/ml +0.075mg/ml +0.2mM/L 17 beta penicillin streptomycin pyruvic acid; follicular fluid group: control group group +10%HFF:EGF control group: liquid liquid +2ng/mlEGF. culture after 24-48 hours of discharge the first polar body oocytes by intracytoplasmic sperm injection (ICSI), 16-18h hours after injection, observation of oocytes, fertilization after cultured for 2 to 3 days of embryo transfer (ET), to observe the maturation rate, fertilization rate, cleavage rate, 2 cell, 8 cell, embryo quality the clinical pregnancy rate and implantation rate.
Result
1, the effect of HFF and EGF on the MII, fertilization and cleavage rate of immature oocytes were significantly higher than that of the control group. The cleavage rate of the former group was significantly higher than that of the control group, while the P0.01 rate, MII rate and fertilization rate had no significant difference between the HFF group and the control group, P.
