【摘要】:前言 人们在观察应激对机体免疫功能影响时发现,应激会导致免疫功能低下、自然杀伤细胞活性受到抑制、淋巴细胞总数减少、单核细胞数减少等等。神经内分泌系统对机体免疫功能具有重要的调控作用,而经典神经递质、神经肽则是直接作用于免疫活性细胞和其他参与免疫反应细胞的重要调节物质。以往有学者报道,SP可促进增殖细胞B产生抗体,并刺激单核细胞产生某些细胞因子共同参与免疫调节作用。有文献指出,无论是体外孵育还是体内给药,SP均可增强小肠上皮淋巴细胞中NK细胞的活性。体内给药能增加脾脏细胞的杀伤力。并且Hart R等发现SP能增加人外周血单核样细胞的IgM及干扰素的分泌。以上资料均表明P物质是体内重要的调节性多肽,并且对免疫功能具有调节作用。神经生长因子(nerve growh factor NGF)是一种嗜神经多肽,亦是发现最早和已知最典型的神经营养因子。其作为一种为感觉神经和交感神经元存活分化和功能所必需的神经营养多肽在神经系统的发育过程中起关键性作用。随着研究的发展和深入,科学家们发现神经生长因子对免疫系统及免疫应答的调节也产生重要影响。 NK细胞是机体十分重要的细胞群体,它几乎参与了免疫系统的发生、发展及效应等重要环节的调节过程。目前,对NK细胞的研究远少于T细胞和B细胞,而有关神经-内分泌-免疫网络中NK细胞所担任的角色、功能变化的完整的文献报道甚少。Hellstrand等发现如果NK细胞经过低浓度的肾上腺素预处理,可以提高NK细胞的活性,但如果10~(-6)M浓度的肾上腺素与NK细胞同育,则NK细胞活性会大大下降。因而,NK细胞对应激原的反应是最敏感的。 神经生长因子(NGF)和P物质(SP)同属多肽类物质。众多资料表明,二者对免疫功能有明显的调节作用。但关于二者在体外对人外周血单个核细胞活性的影响均未见相关报道。本实验研究了SP和NGF对人外周血中
