结核分枝杆菌H37Ra免疫小鼠后机体抗结核细胞免疫功能的研究

【摘要】:结核病(Tuberculosis,TB)是由结核分枝杆菌(Mycobacteriumtuberculosis,MTB)所致的以呼吸系统感染为主的慢性传染病,是目前对人类健康威胁最大的传染性疾病。卡介苗(Bacillus CalmetteGuerin,BCG)是目前预防 TB 唯一的疫苗,但其保护效果不稳定(保护率从 0-80%不等)。新的疫苗的研制已刻不容缓。 目的:探讨结核分枝杆菌 H37Ra 在小鼠体内定植的情况,以及诱导机体产生特异性抗结核细胞免疫功能的情况。 方法:1.用 H37Ra 和 BCG 免疫 BALB/c 小鼠,分别在第 15、30、60天检测结核分枝杆菌在小鼠脏器内的定植。2.免疫后第 30、60 天,检测 BALB/c 小鼠脾淋巴细胞受到 PPD 刺激后增殖转化的刺激指数(Stimulation Index,SI)。3. 免疫后第 30、60 天,检测 BALB/c 小鼠脾淋巴细胞受到 PPD 刺激后白介素-2(interleukin-2,IL-2)的表达。4.免疫后第 30、60 天,检测 BALB/c 小鼠脾淋巴细胞受到 PPD 刺激后可溶性白介素 2 受体(Solubile interleukin 2 receptor,sIL-2R)的表达。 结果:1.H37Ra 和 BCG 均能在 BALB/c 小鼠脏器内定植。2.免疫 30、60 天后,H37Ra 组淋巴细胞刺激指数分别为 2.81±0.63 和 2.16±0.52,BCG 组刺激指数分别为 2.23±0.20 和 2.02±0.38,均明显高于未免疫组(1.33±0.17 和 1.47±0.10),结果有显著性差异(P0.05)。3.免疫30、 天后, 60 H37Ra 组脾淋巴细胞 IL-2 的表达分别为 126.88±8.11pg/ml和 91.26±7.29pg/ml,BCG 组脾淋巴细胞 IL-2 的表达分别为 107.15±
[Abstract]:Tuberculosis tuberculosis is caused by Mycobacterium tuberculosis. MTB) is a chronic infectious disease mainly caused by respiratory infection, which is the most dangerous infectious disease to human health at present. BCG vaccine is Bacillus CalmetteGuerin. BCG is the only vaccine to prevent TB, but its protective effect is unstable (the protection rate ranges from 0 to 80%). The development of new vaccine is urgent. Aim: to investigate the colonization of Mycobacterium tuberculosis H37Ra in mice and the induction of specific anti-tuberculosis cell immune function. Methods BALB/c mice were immunized with H37Ra and BCG respectively at 1530. 60 days after immunization, the colonization of Mycobacterium tuberculosis in mouse viscera was detected. The stimulating index of proliferation and transformation of spleen lymphocytes stimulated by PPD in BALB/c mice was determined.
